With the development of technology,breast cancer surgery has also undergone great changes,although since 1990,the mortality rate of breast cancer has dropped significantly.However,breast cancer still ranks among the top three female morbidity in the world,and the incidence of breast cancer in China is gradually increasing.In this case,it is essential to determine the overall pathological condition of the patient to further determine the surgical and post-treatment options.In this step,the aim of this study is to investigate possible cytokines to assess the staging of patients' pathophysiology in multiple ways.Sox2 and Sox4 belong to the Sox gene family,which are genes composed of SRYrelated genes.The Sox gene family is involved in almost all physiological developments(including sex determination and cartilage development)during the growth and development of individuals.In recent years,some researchers have found that the abnormal expression of Sox gene may be related to the occurrence and development of breast cancer.The Sox2 gene has been confirmed to be abnormally expressed in early breast cancer tissues,because it can participate in the induction of stem cell-like cancer cells.Therefore,it may be one of the factors involved in the early events of breast cancer;the Sox4 gene belongs to the C subfamily of the Sox gene family,and its encoded protein plays an important role in embryonic development and differentiation mainly through the combination of HMG box and DNA.In addition,SOX4 detects a difference table in a variety of cancers,and the function of the Sox4 gene changes as the type of cancer changes.At the same time,the molecular mechanism of SOX4 gene carcinogenesis has not been studied clearly.At present,it has been found that Sox4 can mediate tumor angiogenesis and mediate the occurrence of breast cancer epithelial-mesenchymal transition.PSCA is a 123 amino acid protein expressed on the surface of prostate epithelial cells.At first,only the expression level of PSCA may be closely related to the occurrence and progression of prostate cancer,and it is also one of the important biological indicators for the diagnosis and prognosis of prostate cancer.In recent years,studies have found that PSCA is not only expressed in the prostate,but also in other epithelial malignancies,such as bladder cancer or pancreatic cancer.However,although PSCA expression is expressed in breast cancer tissues,its correlation with clinical factors in breast cancer patients is not known.Therefore,it is speculated that the above cytokines may play a role in breast cancer expression and lymph node metastasis.This article will further verify the possible roles of the above three genes and their expressed proteins.Objective: To analyze the relationship between three proteins and clinicopathological factors of breast cancer by observing the expression of tumor tissue,sentinel lymph node tissue,non-sentinel lymph node tissue and normal breast tissue in breast cancer patients,and to study its relationship with lymph node metastasis.The relationship and further explore the possible role of each protein in the pathological changes of breast cancer.Methods: The expression of breast cancer tissue and normal breast tissue was detected by Real-Time PCR and Western-blot.The two-step immunohistochemistry method was used to detect breast cancer tissue,sentinel lymph node tissue,non-sentinel lymph node tissue and normal breast tissue.Expression,the relationship between the expression of the three cancer tissues and the patient's age,tumor size,histological grade,sentinel lymph node metastasis,non-sentinel lymph node metastasis and epithelial cadherin;statistical analysis of the three sentinel sentinel lymph nodes Statistical analysis was performed on the expression of sentinel lymph nodes with no metastasis;statistical analysis was performed on the expression of non-sentinel lymph nodes and metastasis of non-sentinel lymph nodes with no metastasis;Results: Through RT-PCR,Western-blot and immunohistochemistry,Sox2,Sox4 and PSCA were expressed in normal breast tissue and breast cancer tissues,and the expression level of breast cancer tissues was significantly higher than that of normal breast tissues(p<< 0.01);The positive expression rates of Sox2,Sox4 and PSCA proteins in 2.66 breast cancer tissues were 69.7%,54.5%,and 48.5%,respectively.Three of them were associated with vascular tumor thrombosis,sentinel lymph node metastasis,and non-sentinel lymph node metastasis.Significantly correlated(p<0.05),Sox2 and Sox4 protein expression was significantly associated with E-cad expression(p<0.05),and statistically significant,in terms of age,histological grade,tumor size,ER,PR,and neurological invasion,There was no significant difference in protein between normal and normal breast tissues(p>0.05).3.SOX2 was positively correlated with SOX4 and PSCA proteins(r=0.401,0.453),and Sox4 was positively correlated with PSCA(r).=0.590);4.The relative expression of Sox2,Sox4,and PSCA protein in positive sentinel and non-sentinel lymph nodes was significantly higher than that in negative sentinel and non-sentinel lymph nodes(p<0.05).Conclusion: 1.Sox2,Sox4,PSCA protein are highly expressed in breast cancer tissues and positive lymph nodes,which may become a new index of breast cancer staging.2.Sox2,Sox4,PSCA protein are significantly associated with lymph node metastasis,or can be axillary score The new factor;3.The three are positively related,or play a synergistic role in the occurrence and development of breast cancer.The combined evaluation of the three may become another criterion for individualized treatment.
