Effect Of Podophyllin On The Proliferation Of Human Tongue Cancer Cell Line Tca8113

ObjectiveBy studying the effect of picrodophyllin on the proliferation of tongue cancer cell line Tca8113 and its mechanism,it provides a theoretical basis for the clinical application of picropodophyllin in the treatment of tongue cancer.MethodsThe Tca8113 cells were treated with 0.5,1,2,4,8 ?mol/L PPP and cultured for 24 h,48 h,and 72 h,respectively.The proliferation of Tca8113 cells was detected by MTT assay.The Tca8113 cells were treated with 8 ?mol/L PPP.At 24 h,Hoechst33342 staining was used to observe the morphological changes of cells.Flow cytometry was used to detect the cell cycle of Tca8113 after treatment with 0.5,1,2,4?mol/LPPP for 24 h.Western blotting was used to detect PPP on Tca8113 cells Bcl-2,Bax and Caspase-3 The effect of protein expression;statistical analysis was performed using SPSS 19.0.ResultsThe inhibitory effect of PPP on the proliferation of Tca8113 cells increased with the increase of drug concentration and time,which showed a certain dependence of concentration and time.After staining with Hoechst33342,the treated cytoplasm of Tca8113 was concentrated,and the nucleus was broken and apoptotic.After 24 hours of treatment with PPP,Tca8113 cells showed significant changes in cell cycle,cells in S phase decreased,cells in G2/M phase increased,and the inhibition was concentration-dependent.Western blotting showed that PPP down-regulated Bcl-2 expression and up-regulated Bax and Caspase-3 expression.ConclusionsPPP may inhibit the proliferation of Tca8113 by down-regulating Bcl-2 in Tca8113 and up-regulating the expression of Bax and Caspase-3 in S phase and G2 to M phase,and it may be time-and concentration-dependent.
