Objective:To investigate the effects of SP600125,a JNK inhibitor,on proliferation and migration of human tongue squamous calcinoma Tca8113 cells and explore its machanism.Methods:Human tongue squamous calcinoma Tca8113 cells were treated with 10,20,40?mol/L of JNK inhibitor SP600125 for 24,48,72h.The proliferation ability were investigated by MTT assay.The migration ability were investigated by wound-healing test.Human tongue squamous calcinoma Tca8113 cells were treated with 10,20,40?mol/L of JNK inhibitors SP600125 for 48h.The intracellular localization of JNK and p-JNK in Tca8113 cells was detected by immuno-fluorescence staining.The expression level changes of JNK and p-JNK were detected by Western Blot.Results:1.MTT assay:Tca8113 cells were treated with different concentration of SP600125 ranging from 10,20,40?mol/L for 24,48,72h.The proliferation of Tca8013 cells were significantly inhibited by treatment of SP600125 compared with control group(P<0.01).The results showed that SP600125 inhibit the proliferation of Tca8113 which showed concentration and time dependent manner.2.Wound healing assay:Tca8113 cells were allowed to scratch and migrate for 24,48,72h after treatment with 10,20,40?/L of SP600125.Images of migratory cells from the scratched boundary were observed and acquired with light microscope.The migration of Tca8113 cells were significantly inhibited by treatment of SP600125 for the same concentration compared with control group(P<0.01).The migration of Tca8113 cells were significantly inhibited by treatment of SP600125 for the same time compared with control group.The results showed that SP600125 inhibit the migration of Tca8113 which showed concentration and time dependent manner.3.Immuno-fluorescence staing:Tca8113 cells were treated with different concentration of SP600125 ranging from 10,20,40?mol/L for 48h.JNK was mainly expressed in the cytoplasm,it seems that there was no obvious change in the expression of JNK in the Tca8113 for control and SP600125-treated groups;p-JNK was mainly expressed in the cytoplasm and nuclear,compared with control group,the fluorescence density was reduced in those SP600125-treated group with the increase of drug concentration.4.Western Blot:Tca8113 cells were treated 10,20,40?mol/L of SP600125 for 48h,the expression of p-JNK protein were decreased than the control group(P<0.01),which showed concentration dependent manner.Conclusion:SP600125 significantly inhibit the proliferation and migration of human tongue squamous calcinoma Tca8113 cells.which showed dose dependent manner.
