Objective:This paper aims to study the anti-tumor activity of Coenzyme Q10 on liver cancer,sarcoma,gastric cancer,colorectal cancer and glioma by constructing animal models,and then to study the anti-tumor effect of coenzyme Q10 combined with artemether on glioma and colorectal cancer to explore its synergistic characteristics.In summary,this paper aims to provide a theoretical basis for the future application of artemether and coenzyme Q10 in the clinical field.Method:1.The tumor cells were inoculated in the subcutaneous position of the right forelimb of mice to construct a subcutaneous xenograft model of H22 liver cancer,sarcoma and colorectal cancer,and a subcutaneous xenograft model of human MKN-28 gastric cancer.An SD animal model of mouse orthotopic glioma was constructed using SD rats as experimental subjects.The data on the antitumor properties and inhibitory activities of coenzyme Q10 and artemether in animals were obtained by intragastric administration of the experimental mice after successful modeling.2.Using HE staining,the changes of tumor cells in subcutaneous colorectal cancer tissues of BALB/c mice were detected after coenzyme Q10 and artemether administration.3.Using immunohistochemistiy to detect the characteristics of tumor angiogenesis in glioma tissues.Results:1.In the case of coenzyme Q10,compared with the negative control group,the inhibition rate of the experimental group on liver cancer,sarcoma,gastric cancer,colorectal cancer and glioma was 76.47%(P<0.01),65.15%(P<0.01),61.28%(P<0.05),38.97%(P>0.05)and 15.3%(P>0.05),indicating that coenzyme Q10 has significant inhibitory effects on liver cancer,sarcoma and gastric cancer,and has certain inhibitory activity on the growth of colorectal cancer and glioma.2.In the case of artemether.compared with the negative control group,the inhibition rate of the experimental group on colorectal cancer and glioma was 85.47%(P<0.05)and 52%(P<0.05),respectively,indicating that artemether has significant inhibitory effects on colorectal cancer and glioma.3.Artemether combined with coenzyme Q10,compared with the negative control group,the inhibition rate of the experimental group on colorectal cancer and glioma was 26.06%(P>0.05)and 23.7%(P>0.05),respectively,both lower than the artemether alone administration group,indicating that the combination of coenzyme Q10 and artemether did not have a positive synergistic effect on the inhibition of colorectal cancer and glioma.4.HE staining showed that the colorectal cancer cells in the negative control group were heavily stained,densely arranged,numerous in number,and the number of tumor cells with nuclear fission was high.The number of colorectal cancer cells in the artemether administration group was significantly reduced,and the tumor cells were only dispersed,and the number of tumor cells with nuclear fission was extremely small,indicating that artemether has an inhibitory effect on the growth of colorectal cancer cells.5.Immunohistochemistry showed that compared with the negative control group,the number of tumor angiogenesis in glioma tissues of artemether administration group was significantly reduced,indicating that artemether has an inhibitory effect on angiogenesis of glioma.Conclusion:The results of in vivo experiments confirmed that coenzyme Q10 has inhibitory effects on liver cancer,sarcoma,gastric cancer,glioma and colorectal cancer,among which,the inhibitory effect on liver cancer,sarcoma and gastric cancer is significant.Artemether has a significant inhibitory effect on glioma and colorectal cancer.At the same time,the experimental results show that Coenzyme Q10 and artemether do not have a positive synergistic effect in inhibiting tumor growth.In addition,from the results of immunohistochemistry,the inhibitory effect of artemether on glioma is achieved by inhibiting the formation of tumor blood vessels.
