Comparative Research On Efficacy Of Co-artemether And Co-artemether Coordinate With Astragalus Treating Artemisinin-Resistant Plasmodium Berghei Rodent Malaria And Chloroquine Resistant Rodent Malaria

Objective:The establishment of artemisinin-resistant Plasmodium berghei rodent malaria model, further study of the efficacy differences of co-artemether, co-artemether compound prescriptions of traditional Chinese medicine Astragalus and other antimalarial drugs treating Artemisinin-resistant Plasmodium berghei rodent malaria and Chloroquine resistant rodent malaria differences based on the previous experiment.Methods:Based on the previous artemisinin-resistant Plasmodium berghei strains cultivation, continue to four days of drugs inhibition and gradually increase the dose of every generation, and measure the blood smear negative conversion rate and ED50 by tail vein, and then calculate the resistance index of the latest resistant strains. To set up the equivalent dose group of co-artemether, artemether, artemisinin, lumefantrine, and Huang Qi according to the Plasmodium berghei normal rodent malaria model, to conversion the equivalent treatment dose of Huang Qi in animal with the conversion coefficient of body weight dose of animal and human per kilogram to on the drug, analyze the difference of dose effect of co-artemether in 1 times,1.5 times,2 times, 2.5 times and 3 times of the equivalent treating the artemisinin resistant model of Plasmodium berghei malaria, and also the efficacy difference while compound with Huang Qi; differences in efficacy of co-artemether, artemisinin, artemether, lumefantrine and Huang Qi against Plasmodium berghei chloroquine resistant rodent malaria. The anti-malarial efficacy experiment using four days of drugs inhibition, measure the parasitemia densities through blood smear 24h after taking the drugs, cure rate of 28 days and conversion rate.Results:1.The amount of artemisinin given to plasmodium berghei normal strain was increased gradually to 27 generation, study the resistance index of the 24 generation, the ED50 reaches 2098.575 mg/kg, and Resistance index comes to 8.31.2. Inhibitory effect can be observed while giving co-artemether 1 times the equivalent dose and also 1.5 times,2 times,2.5 times,3 times the equivalent dose of co-artemether to artemisinin resistant rodent malaria of Plasmodium berghei, however none of the group has 100% cure rate in 28days, The negative conversion ratio between the groups was statistically significant. The co-artemether is observed suspected resistance when treating artemisinin resistant rodent malaria of Plasmodium berghei (Parasitemia densities> 0.005).3. Huang Qi has no directly effect on the treatment of Artemisinin-resistant Plasmodium berghei rodent malaria and Chloroquine resistant rodent malaria, but when the treatment dose of co-artemether was 3 times the standard dose, combined with Huang Qi can improve the effect of drug treatment(P<0.05). By comparing of the average weight change, mortality in 28 days and coefficient of liver and spleen between the groups, combined with Huang Qi can reduce the body weight decreased, decreased liver and spleen coefficient and prolong life, Improve the quality of life of mice. However no reversed efficacy can be observed in the liver and spleen after been injured in mice when combined with Huang Qi in the treatment groups.4. The curative rate of co-artemether and combined with Huang Qi in the treatment of chloroquine resistant Plasmodium berghei rodent malaria can up to 100% in 28 days, while the curative rate of artemether, lumefantrine alone and combined with Huang Qi in the treatment of chloroquine resistant Plasmodium berghei rodent malaria was less than 100%. Curative effect showed no significant difference when the co-artemether combined with Huang Qi(P>0.05), while the curative rate improved when artemether and lumefantrine, combined with Huang Qi(P<0.05).5. Experimental observation of Plasmodium berghei chloroquine resistant rodent malaria shows a certain self-healing rate. References suspect that mouse self-healing performance may be related to a virulent weaken of chloroquine resistant strain.Conclusion:The experimental results show that the Plasmodium berghei artemisinin resistant rodent malaria model is establishing, the resistance index comes to 8.31; the co-artemether is observed suspected resistance when treating artemisinin resistant rodent malaria of Plasmodium berghei, the cultivation rate can not up to 100% though the doses of co-artemether are increased; co-artemether has better curative effect compared with artemether and lumefantrine 24h after taking the drugs, and the curative effect can be improved when combined with Huang Qi.
