Research background and purposeHemorrhoids is located in the anus of a kind of common disease,relevant pathological studies of hemorrhoids is less,its molecular mechanism is unclear,and VEGF and FGF2 has important significance in promoting hemorrhoids blood vessel growth,this study by histopathology,immunohistochemistry and western blot method to observe the human body in different stages of hemorrhoids mucosa and vascular the expression of VEGF and FGF2,investigate the occurrence and development mechanism of hemorrhoids,to provide theoretical basis for the prevention and treatment of hemorrhoids.Research data and methodsCollected from nanfang hospital,southern medical university in March 2017-March 2018 specimens of anorectal outpatient hemorrhoids resection specimen of 134 cases,including ? period,42 cases ? period 45 cases,? 47,and hemorrhoids around 40 cases of normal intestinal wall tissues as control;Another collecting fresh hemorrhoid with the 15 cases in men,women,? period 10 cases,? period of 10 cases,10 cases ? period.The mucosal layer and vascular pathology of hemorrhoid tissue were observed by HE staining.The expression and distribution of VEGF and FGF2 in hemorrhoid mucosa and blood vessels were detected by immunohistochemistry.Protein expression levels of VEGF and FGF2 in hemorrhoid mucosa and blood vessels were detected by western blotting.SPSS 13.0 software was used for statistical analysis.Anova was used for multigroup mean comparison,and SNK test was used for pantwise comparison.Test level =0.05.RESULTSPathology observation group and normal phase ? hemorrhoids mucosa covering epithelium is complete,not seen to dilate blood vessels;? hemorrhoids mucosa covering epithelium damage,mucosal muscularis,mucosa and mucosal muscularis is visible within the new angiogenesis;Phase ? hemorrhoids mucosa covering epithelium damage increase,see blood vessel wall form circuity,tube cavity expansion.Immunohistochemistry showed that,compared with normal mucosal fibroblasts,the expression level of VEGF(F=883.961,P<0.01)and FGF2(F=656.013,P<0.01)in mucosal fibroblasts increased with the increase of clinical stages.Compared with the normal group,the expression level of VEGF(F=776.561,P<0.01)and FGF2(F=1066.458,P<0.01)in vascular endothelial cells increased with the increase of clinical stages.Westerm blot test results showed that the protein expression levels of VEGF(F=65.312,P<0.01)and FGF2(F=129.125,P<0.01)in hemorrhoid tissues at each stage increased with the clinical stage of internal hemorrhoids.CONCLUSIONSTVEGF,FGF2 in hemorrhoids vascular endothelial cells and the expression level of fibroblasts in mucous membrane layer expression levels with lesions is aggravating,presumably in the process of occurrence and development of hemorrhoids VEGF,FGF2 have not only promoted the hemorrhoids vascular epithelial hyperplasia,and also promoted the hemorrhoids into fiber cell proliferation and the expression of VEGF and FGF2 level was positively correlated with the progression of hemorrhoids,the important molecular markers can be used as the mechanism of hemorrhoids.
