Purpose:Through detecting the expression of ferritin heavy chain/ferritin light chain and c-kit in epithelial ovarian cancer tissues,we analyzed the correlation between these three proteins as well as the relationship between FHC/FLC expression and clinicopathological characteristics of patients with c-kit positive epithelial ovarian cancer.To investigate the role and clinical significance of ferritin heavy chain and light chain in c-kit positive ovarian cancer tissues.Method:A total of 159 patients with epithelial ovarian cancer who were treated at the First Affiliated Hospital of Yangtze University from 2011 to 2018 were collected.40 cases of normal ovarian tissue.All patients had postoperative pathological diagnosis confirmed as epithelial ovarian cancer.The expression of c-kit,FHC and FLC in 199 tissues was detected by immunohistochemistry.The experimental data were processed by SPSS software,and the expression was compared with the clinical data of clinical stage and pathological grade of epithelial ovarian cancer.Results:1.Compared with normal ovarian tissue,FHC has a higher expression rate in EOC tissues(32.5%vs 70.00%),and the difference is statistically significant(x~2=13.64;P<0.01);FLC has a higher expression rate in EOC organization(25.00%vs 63.33%),and the difference was statistically significant(x~2=14.13;P<0.01).2.There was no significant difference in c-kit with age and tumor type(P>0.05).In EOC tissues with FIGO stage I-II and III-IV,the positive expression rates of c-kit were12.94%and 28.37%,respectively.the difference between the two groups was statistically significant(x~2=5.86;P=0.015).In the EOC tissues of the pathological grade,the positive rates of c-kit expression were 12.31%and 36.17%,respectively,and the difference between the two groups was statistically significant(x~2=4.18;P=0.04).3.There was no significant difference in FHC between FIGO stage,age and tumor type(P>0.05).In pathological grade,the positive rate of FHC expression was 74.47%and 58.46%,respectively.and the difference between the two groups was statistically significant(x~2=4.52;P=0.03).4.There was no significant difference in FLC between EOC and age and tumor type(P>0.05).In FIGO stage,the positive expression rate of FLC was 55.29%and 78.38%,respectively.The difference between the two groups was statistically significant(x~2=9.40;P=0.002).In the pathological grade,the positive rates of FLC expression were68.09%and 41.54%,respectively,and the difference between the two groups was statistically significant(x~2=11.06;P<0.01).5.The positive rates of FHC expression in c-kit+EOC tissues and c-kit-EOC tissues were 56.25%and 70.86%,respectively.There was a negative correlation between FHC and c-kit expression,and the correlation coefficient was r=-0.23.The difference between the two was not statistically significant(x~2=2.5;P=0.11);The positive rates of FLC expression in c-kit+EOC tissues and c-kit-EOC tissues were 43.75%and 70.08%,respectively,and the difference was statistically significant(x~2=7.76;P<0.05).6.There was no significant difference between FLC expression and FIGO clinical stage and pathological grade in c-kit+EOC tissues(P>0.05).There was no significant difference between FHC expression and pathological grade(P>0.05).FHC expression in FIGO clinical stage was the expression rates of I-II and III-IV were 27.27%and 71.43%,respectively,and the difference was statistically significant(P<0.05).Conclusion:1.Compared with normal ovarian tissue,FHC and FLC are highly expressed in epithelial ovarian cancer.In EOC tissues,FHC and FLC were positively correlated with tissue grade and FIGO stage,suggesting that ferritin may be one of the markers for diagnosis and prognosis of ovarian cancer.2.The expression of FLC in c-kit positive epithelial ovarian cancer tissues was lower than that in c-kit negative tissues.It was also found that FHC expression was positively correlated with FIGO stage,suggesting that the ratio of ferritin heavy chain and light chain in cancer stem cell marker c-kit tissue or There is a change in structure,and FHC may be associated with markers of ovarian cancer metastasis and prognosis.3.Abnormal expression of ferritin heavy chain may be one of the characteristics of c-kit positive epithelial ovarian cancer,which may provide new ideas for the diagnosis and treatment of ovarian cancer.
