The Bitong Miture For Research On The Effects Of Type Of Adjuvant Arthritis Model In Rats

Objective:To investigate the pharmacodynamic effects of Bitong Mixture on adjuvant arthritis model rats.Method:Forty SD rats were randomly divided into five groups:blank group,model group,diclofenac sodium group,Bitong mixture high-dose group(high-dose group),Bitong mixture low-dose group(low-dose group).Except the normal group,the other rats were injected with 0.2 ml Freund's complete adjuvant intradermally into the right hind toe to form aduvant-induced arthritis rat model.The normal group was injected with saline of equal volume at the same place.After inflammation,rats in each group were given continuous intragastric administration for 21 days according to the set dose.The daily dose of diclofenac sodium group was 0.5mg/100g diclofenac sodium,the daily dose of Bitong mixture in high dose group was 0.6ml/100g Bitong mixture,and the daily dose of Bitong mixture in low dose group was 0.3ml/100g Bitong mixture.Distilled water of equal volume was given to blank group and model group respectively.The histopathological changes of synovium,the expression levels of inflammatory factors IL-1beta,IL-6 and TNF-alpha,and the relative expression levels of RIP140/NF-kappa B signaling pathway related proteins were detected.Result:1.Compared with the model group,the positive group and the low-dose group,the high-dose group of Bitong Mixture increased the weight of rats(P<0.05).2.Compared with the model group,the positive group and the low-dose group,the high-dose group of Bitong Mixture decreased the toe volume of rats(P<0.05).3.Compared with the model group,the positive drug group and the low-dose Bitong mixture group,the high-dose Bitong mixture group had less infiltration of inflammatory cells on the cartilage surface and clearer structure.4.Compared with the model group,the inflammatory cytokines such as IL-1beta,IL-6 and TNF-alpha in synovial tissue of the positive drug group,Bitong mixture low-dose group and Bitong mixture high-dose group were significantly lower(P<0.05).5.After treatment,the expression of IKBa/p-I?B??P-P65/P65?MyD88?TLR4 and RIP 140 protein in rats was significantly decreased in low-dose Bitong mixture group and high-dose Bitong mixture group.Conclusion:Bitong Mixture can inhibit secondary joint swelling,significantly reduce the levels of IL-lbeta,IL-6 and TNF-alpha in serum of adjuvant arthritis rats,and inhibit RIP140/NF-kappa B signaling pathway in synovial tissue.In a word,Bitong Mixture high dose group is better than low dose group.
