Objective:Gastrodia eata Gastrodia eata is the first of the "ten authentic medicinal materials" in Guizhou.It is a traditional and precious Chinese medicine.It has a medicinal history of more than two thousand years,and has the effect of calming the liver and relieving the wind and dispelling wind and collateral.But the study of different size Gastrodia elata powder on cerebral ischemia-reperfusion and sedative analgesia is not comprehensive.On the basis of clarifying the effectiveness of Gastrodia elata in the treatment of cerebral ischemia-reperfusion and sedative analgesia,this study investigated the difference of Gastrodia elata under different particle sizes and clarified the correlation between particle size and pharmacological effect.The optimum particle size of Gastrodia elata powder was selected from the aspect of pharmacodynamics,and the scientific nature,rationality and safety of ultrafine Gastrodia elata powder were discussed from the level of pharmacodynamics,which provided a more instructive significance for the application of ultrafine pulverizing technology in the modernization of traditional Chinese medicine.Methods;(1)The model of focal ischemia-reperfasion injury(Middle Cerebral Artery Occlusion,MCAO)was prepared by single side thread embolization.The rats were divided into sham-operation group,model group,ultrafine Gastrodia elata powder group ?,? group,? group,positive drug group.The activity of superoxide dismutase(Superoxide dismutase,SOD),the content of malondialdehyde(Malondialdehyde,MDA),the activity of nitric oxide synthase(Nitricoxide synthase,NOS),and the pathological changes of brain tissue were compared in each group.Evaluation of the efficacy of ultrafine Gastrodia elata powder with different particle sizes in the treatment of cerebral ischemia-reperfusion injury.(2)The simultaneous determination of glutamic acid(Glutamic acid,Glu)and aspartic acid(Aspartic asp),glycine(Glycine,Gly)in brain homogenate was established by high liquid chromatography-electrospray tandem mass spectrometry(HPLC-ESI-MS/MS).Five neurotransmitters of 5-hydroxytryptamine(5-hydroxytryptaphane 5-HT)and valine(Valine Val)were used to study the effects of oral administration of different diameters of Gastrodia elata ultrafine powder on the content of five neurotransmitters in each group.The differences of five neurotransmitters in each group were compared and analyzed.(3)The analgesic effects of five different sizes of Gastrodia elata powder were observed by acetic acid writhing method and hot plate method in mice.The effects of different particle sizes of Gastrodia elata powder on sedation of mice were observed by intraperitoneal injection of pentobarbital sodium.Results:Compared with the sham operation group,the model group showed obvious signs of neurobehavioral deficit(P<0.01),and the area of cerebral infarction increased significantly(P<0.01).Compared with the model group,the ultrafine Gastrodia elata powder iiig ? group significantly improved the neurobehavioral function(P<0.05),significantly decreased the area ratio of cerebral infarction,the content of glu,asp,val,mda and the activity of nos(P<0.05 or P<0.01).The activity of sod and the content of gly and 5-ht were significantly increased(P<0.05).The activity of sod and the content of gly in ? group were significantly higher than those in ? group and ? group(P<0.05).The content of mda and the activity of nos were significantly lower in group ? and ii than those in group? and ?(O<0.05).Compared with the blank group,the duration of sleep in each group increased significantly(O<0.05),and the duration of sleep in the 400 mesh group of Gastrodia elata was higher than that in the other groups,indicating that all five kinds of Gastrodia elata powder had a synergistic effect on pentobarbital sodium.Compared with the blank group,the times of writhing in mice induced by acetic acid were significantly decreased in each group(P<0.05),and the times of writhing in 400 mesh and 500 mesh were significantly decreased(P<0.05)compared with 100 mesh of Gastrodia elata powder.The inhibitory rate was also increased to some extent,indicating that all five kinds of Gastrodia elata powder had analgesic effect on mice,and 400 and 500 meshes had stronger effects.Conclusion:Gastrodia elata powder with different particle size had significant protective effect on cerebral ischemia-reperfusion injury and affected the above indexes from different levels.But the preventive and protective effect of Gastrodia elata ultrafine powder iii,ii was more obvious than that of Gastrodia elata ultrafine powder.Different particle sizes of Gastrodia eata powder could obviously enhance the sedative and analgesic effect on mice,and the effect of Gastrodia eata ultrafine powder? was more remarkable.It can be seen that the particle size has some effect on the preventive and protective effect of Gastrodia elata,which may be due to the finer the particle size of Gastrodia elata powder.The results showed that the cell wall was broken more thoroughly,the active components of Gastrodia elata were fully exposed,it was more favorable to its dissolution and absorption,and the bioavailability was improved,so that the drug effect could be brought into full play.In this study,the results of the two aspects of Gastrodia elata showed that ?,? had a strong effect,which provided a reference for the selection of the best particle size of Gastrodia elata.
