Objective: By analyzing the immunohistochemical characteristics of CD86,CD163 and CD1 a in the infected tissues of Talaromycosis marneffei(TM),it was discussed whether the positive immunohistochemistry(IHC)staining of the yeast-like cells was an accidental or common phenomenon.Methods: The samples comprise 12 paraffin-embedded skin tissues from HIV(+)patients infected by TM,3 paraffin-embedded skin tissues from HIV(-)patients infected by TM,1 paraffin-embedded liver tissue and 1paraffin-embedded lung tissue from mice infected by TM,1 paraffin-embedded cell mass from the yeast-like cells of TM were co-cultured with macrophages,and 1 paraffin-embedded cell mass from the yeast-like cells were cultured alone.The tissue slides were prepared to compare the staining differences of CD86,CD163 and CD1 a in the tissues infected by TM in vivo and in vitro by IHC staining.Results:1.HIV(+)patients infected by TM:(1)The yeast-like cells of TM are diffusely distributed,a large number of cells are released outside the cell.CD86 staining showed high intensity positivity in the yeast-like cells of TM,CD163 and CD1a staining showed a negative result in the yeast-like cells of TM;(2)The expression of CD86 was scattered and not obvious in histiocytes,but CD163 was obviously positive in histiocytes.(3)The expression of CD1 a was positive in Langerhans cells.2.HIV(+)patients infected by TM:(1)The number of yeast-like cells of TM was small and scattered,positive for CD86 staining,negative for CD163 and CD1 a staining;(2)The positive expression of CD86 in histiocytes was enhanced compared with that in HIV(+)TM patients,and CD163 was positively expressed.3.The animal models infected by TM:(1)The yeast-like cells of TM were strongly positive for CD86 staining,and negative for CD163 and CD1 a staining;(2)Both histiocytes and multinucleated giant cells had positive expression of CD86 and CD163.4.The yeast-like cells of TM were co-cultured with macrophages:(1)The yeast-like cells of TM were strongly positive for CD86 staining,and negative for CD163 and CD1 a staining;(2)Strong positive expression of CD86 in macrophages.5.The yeast-like cells of TM were cultured alone: The yeast-like cells of TM were strongly positive for CD86 staining,and negative for CD163 and CD1 a staining.Conclusion:The yeast-like cells of TM may produce CD86-like glycoproteins.
