Objective To observe the expression of Wnt3a,?-catenin and CyclinD1 in hippocampus at different time after diffuse brain injury,and to explore the mechanism of Wnt/?-catenin signaling pathway in brain injury and its relationship with the time of injury.It provides experimental basis for forensic identification and molecular pathological study of diffuse brain injury.Methods Thirty healthy adult SD rats weighing 300±10g were randomly divided into six groups:normal control group,DBI 6h group,DBI 1d group,DBI 2d group,DBI 5d group and DBI 7d group,with 5 rats in each group.Establishment of DBI model according to Marmarou's principle of free falling body striking method.To observe the behavioral changes of rats after diffuse brain injury,and to observe the pathological changes of hippocampus by H-E staining.The expression and change of Wnt3a,?-catenin,CyclinD1 gene of Wnt/?-catenin signaling pathway in hippocampus were detected by immunohistochemical staining and Western Blot.Results Wnt3a,?-catenin and CyclinD1 were expressed in a small amount in hippocampus of normal control group rats.Compared with the normal control group,the expression of Wnt3a in hippocampus of the model group began to increase 6 hours after injury,peaked on the 1st day,gradually decreased on the 2nd day,and increased on the 7th day after injury.The expression of?-catenin began to increase 6 hours after injury,peaked on the 1st day,then gradually decreased,and fell to normal level on the7th day after injury.The expression of CyclinD1 began to increase sharply at 6 hours after injury and reached the peak,then decreased gradually after the 1st day,and dropped to normal level on the 7th day after injury.Conclusion Wnt/?-catenin signaling pathway plays an important role in the regulation of diffuse brain injury,which may be mediated by the activation and expression of the signal to promote the proliferation of neurons in the hippocampus after diffuse brain injury,in order to complete the regeneration and repair of the injured nerve;The expression of Wnt3a,?-catenin and CyclinD1 genes is different in time,which may be used as an index for predicting the time of early injury.It has certain guiding significance for the study of molecular pathological mechanism of diffuse brain injury and forensic identification of brain injury.
