Purpose:According to the records of Chinese medicine classics,the roots of the platycodon grandiflorum,such as Codonopsis pilosula,Chuan Dangshen or Codonopsis,are collectively referred to as medicinal Codonopsis.Dangshen is flat and sweet,has the effect of replenishing the spleen,lungs,nourishing the blood and fluid,and is a medicine for qi.For the treatment of Codonopsis pilosula with lung qi deficiency,asthma cough and spontaneous sweat,qi and blood deficiency,gas and Jin two injuries,limbs fatigue and fatigue syndrome have a very good effect.In recent years,with the development of traditional Chinese medicine encephalopathy,there has been a class of spleen-stomach debilitating encephalopathy in Chinese medicine,mainly characterized by memory loss,insomnia,dizziness,insufficient blood supply to the brain,etc.In the traditional Chinese medicine prescription for treating such diseases,Codonopsis pilosula has been applied.The dosage of this kind of medicine is relatively large.The effect of Codonopsis pilosula in the compound is to replenish qi and invigorate the spleen and to replenish the blood.Then,does the Codonopsis pilosula single-medicine have the effect of nourishing the brain,whether it passes the spleen and stomach,and qi and nourish the blood.The mechanism to improve brain function? Method:The study consists of three parts.The first part is the experiment of improving the memory function of mice with different extracts of Codonopsis pilosula.The model of memory impairment in mice was established by scopolamine hydrobromide.The spatial memory ability of mice was passive using Morris water maze and dark evaluator.Avoid memory ability to detect and evaluate the effect.Then,DTNB spectrophotometric method was used to detect the levels of glutathione catalase(GSH-PX),malondialdehyde(MDA)and superoxide dismutase(SOD)in the serum of mice.The second part is the experiment of codonopsis pilosula extract to enhance the gastrointestinal function of mice: the gastrointestinal motility of mice is detected by calculating the small intestine propulsion rate,and the relative residual rate in the stomach is measured by spectrophotometer at 620 nm,and finally the radioimmunoassay is used.The method measures the values of gastrin(GAS)and motilin(MTL)in serum.Cisapride is a positive control for this experiment.The third part is the experiment of increasing the hematopoietic capacity of the codonopsis pilosula extract: the value of red blood cells,hemoglobin and hematocrit is detected by automatic blood cell analyzer.Angelica is the positive control for this experiment.Result:1)The mice of the Codonopsis pilosula extract group had a shorter time to find the platform hidden under the water surface,and the incubation period was shorter;the Dangshen water extract group had a longer time and the incubation period was prolonged.The number of times of platform and platform stay time of Dangshen alcohol extract group increased.The number of platform and platform stay time of Dangshen water extract group increased significantly.There was no significant change in the latency and dark-avoidance latency of the mice in the alcohol extract group of Dangshen.The mice in the water extract of Codonopsis pilosula showed better performance,and the latency and avoidance latency of the platform were significantly shortened.The levels of serum GSH-PX and SOD in the serum of Dangshen alcohol extract group increased,and the change of MDA was not obvious.The serum SOD level of Dangshen aqueous extract group was significantly increased,while GSH-PX and MDA did not change significantly.The overall effect of the water extract of Codonopsis pilosula was relatively good.Therefore,the water extract of Codonopsis pilosula was selected for follow-up experimental research.2)The relative residual rate of intragastric pigment in the cisapride group was significantly lower than that in the blank control group,while the Codonopsis group did not change significantly;the small intestine propulsion rate of the cisapride group and the Codonopsis group increased significantly,while the cisapride group and the Codonopsis group There was no significant difference between them.Serum GAS and MTL levels increased in the cisapride group,and MTL levels in the Codonopsis group increased,while GAS levels did not change significantly.3)Both the Angelica group and the Codonopsis group could increase the values of red blood cells,hemoglobin and hematocrit in the serum of mice,and there was no significant difference between the Angelica group and the Dangshen water extract group.In conclusion:In this study,the research on the memory function,gastrointestinal function and blood function of the mice was carried out from the extract of Codonopsis pilosula.The mechanism of the three functions of Codonopsis pilosula was improved.The results show:1)Codonopsis pilosula extract can promote the promotion of intestinal contents and increase the level of gastrointestinal stimulating hormone-motilin.2)Codonopsis extract can increase the value of red blood cells,hemoglobin,hematocrit in blood deficiency mice.3)Codonopsis pilosula extract improves the ability of learning and memory and passive avoidance of memory in mice with memory impairment,and at the same time increases the level of SOD in serum to reduce lipid peroxidation,thereby achieving anti-oxidation and reducing damage of brain tissue by antioxidant substances.
