ObjectiveThis work was to investigate the effects of Panaxjaponicus decoction on learning and memory function and Synaptophysin-related protein expression in hippocampus of Alzheimer's disease model(A?25-355-35 induced)rats from the pathogenesis of deficiency-stagnation-phlegm in traditional Chinese medicine.Methods?1?Theoretical research:Through consulting the literature,reviewing the pathogenesis of AD in classics of traditional Chinese medicine,and combining with the research results,further understanded basis pathogenesis of AD.?2?Experimental study:Sixty SD rats of three months were randomly divided into sham operation group,model group,drug group,panaxJaponicus low dose group,panaxJaponicus medium dose group and panaxJaponicus high dosegroup.The model was induced by A?25-355-35 bilateral hippocampus injection,and the corresponding dose of panaxJaponicus decoction,donepezil hydrochloride and normal saline were administered continuously by gastric perfusion for 28 days.The spatial learning and memory ability of AD rats after 21 days of medication were detected by MWM test.The expression of A?1-42-42 in hippocampus of AD rats was detected by ELISA.The expressions of BACE-1,Synaptophysin,SynapsinI and PSD-95 protein in hippocampus of AD rats were detected by western blot.The expression of PSD-95protein in hippocampal CA1 and CA3 regions of AD rats was detected by IHC.Results?1?Compared with the AD model group,the swimming time of sham-operated control group at the target area decreased significantly?P<0.01?,the number of times the rat passes through the original platform area reduced significantly?P<0.01?,and the time to cross the platform for the first time increased significantly?P<0.01?.Compared with the AD model group,the swimming time of panaxJaponicus therapy group at the target area increased significantly?P<0.05?,the number of times the rat passes through the original platform area upregulated significantly?P<0.05?,and the first time to cross the platform decreased significantly?P<0.05?.No difference was observed between the panaxJaponicus treatment groups and the western medicine group.There was no significant difference in the speed of the whole course between the rats in each group?P>0.05?.?2?Compared with the sham operation group,the A?1-42-42 of hippocampus was increased in the model group by using ELISA?P<0.01?.Compared with the model group,the medium and high dose groups of panaxJaponicus decreased significantly?P<0.05?.The difference between panaxJaponicus therapy group and the western medicine group was not obvious?P>0.05?.?3?The expression of BACE-1,Synaptophysin,SynapsinI and PSD-95 protein in hippocampus was detected by Western blot.Compared with the sham operation group,the BACE-1 was increased in the model group?P<0.01?,and the Synaptophysin,SynapsinI and PSD-95 were decreased?P<0.01?.Compared with the model group,the BACE-1 was significantly decreased in the medium and high dose groups of panaxJaponicus?P<0.01?,and the content of Synaptophysin and SynapsinI were increased in each dose of panaxJaponicus?P<0.05?,the PSD-95protein was significantly increased in the medium and high dose groups of panaxJaponicus?P<0.01?.The difference between panaxJaponicus therapy group and the western medicine group was not obvious?P>0.05?.?4?Compared with the sham operation group,the PSD-95 protein content in the hippocampal CA1 and CA3 areas of the model group decreased significantly?P<0.01?.Compared with the model group,the expression of PSD-95 in the CA1 and CA3 areas increased significantly in panaxJaponicus group?P<0.05?,but no difference was observed between in the high dose groups of panaxJaponicus group and the western medicine group?P>0.05?.Conclusion?1?The deficiency of healthy energy in body and the functional imbalance of liver crgan,spleen organ,kidney organ and so on.The pathological changes of blood stasis and phlegm coagulation blockage in the brain.The above factors lead to marrow-sea defiaiency and mental dysfunction which is an important pathogenesis of AD.?2?The panaxJaponicus decoction can improve the learning and memory function of AD rats induced by A?25-35.?3?PanaxJaponicus decoction can reduce the expression of BACE-1 and A?1-42in hippocampus of AD rats induced by A?25-35.The mechnism is that the formation of A?is reduced through decreasing the cleavage of APP by BACE-1.Meanwhile,PanaxJaponicuspromotes the clearance of A?1-42-42 and reducing the A?to reduce neurotoxic damage.?4?PanaxJaponicus decoction up-regulates the expression of synapsinI,Synaptophysin and PSD-95 in hippocampus of AD rats topromote the remodeling of synaptic structure and function.
