Allium victorialis L.,a medicinal plant,is a perennial herb belonging to Allium,Liliaceae.It contains flavonoids,steroids,sugars and sulfur-containing components.This herb has many functions such as hemostasis,removing phlegm,analgesia and so on.It can be used for cold,nausea,swelling and other diseases.Recently,the biological activities,including anti-inflammatory,anti-obesity,treatment of diabetic nephropathy,bacteriostasis,prevention and treatment of cardiovascular diseases,anti-neuritis,have been reported by domestic and foreign researchers.In order to further elucidate its chemical constituents and to screen its biological activity against chronic obstructive pulmonary disease?COPD?,based on the review of the study on Allium victorialis L.and COPD,the following works about Allium victorialis cultivated in Jilin province had been carried out in this thesis.1.The technology of HS-SPME combined with GC-MS was used to determine and analysis the volatile components of bulbs,stems and leaves,scape and flowers,fibrous root of Allium victorialis for the first time.As a result,a total of 179compounds,including 12 shared components in four morphological regions,were identified.It could be indicated that there were much different in the types and contents of volatile components in four parts of Allium victorialis.Among the identified compounds,the main components were sulfur-containing substances,and the total contents of the four parts were 79.75%,52.77%,59.55%and 47.83%,respectively.2.The chemical constituents of the whole plant were extracted and isolated by repeated silica gel column chromatography,Sephadex LH-20 and semi-preparative HPLC.As a result,18 compounds were isolated from 70%ethanol extract of Allium victorialis?EEAV?.Among these,15 compounds were identified by TLC,HR-MS and NMR as follows:gitogenin?1?,?-sitosterol?2?,xylopyranosetetra-benzoate?3?,n-Butyl-?-D-fructopy ranoside?4?,daucosterol?5?,?22S?-cholest-5-ene-1?,3?,16?,22-tetraol-O-?-L-rhamnopyranosyl-16-O-?-D-glucopyranoside?6?,p-Hydroxypheneth-yl alcohol?7?,quercetin?8?,quercitroside?9?,kaempferide?10?,caffeic acid?11?,cinnamic acid?12?,apigenin?13?,2-?Methoxymethyl?-thiazole?14?and 5-hydroxy-methyl furfural?15?.Among of them,11 compounds including compounds 1,3,4,6,7,11,12,13,14,15 were isolated from Allium victorialis for the first time.3.The anti-COPD activity of Allium victorialis was preliminarily evaluated for the first time by intragastric administration EEAV(0.16 g·kg-1,0.32 g·kg-1)to COPD mice.The detected indexes included lung function,serum tumor necrosis factor-??TNF-??,interleukin-6?IL-6?,interleukin-1??IL-1??,malondialdehyde?MDA?and superoxide dismutase?SOD?levels and lung histopathology.The results showed that the body weight,lung function and the activity of SOD were significantly decreased?p<0.01?,while the contents of TNF-?,IL-1?,IL-6 and MDA were significantly increased?p<0.01?in COPD model group.At the same time,severe lung lesions were observed in model group,indicating that the construction of the COPD model was successful.Compared with the model group,the body weights in EEAV groups were both increased significantly,MDA level were both decreased significantly?p<0.05?.While in high dosage EEAV group,FEV0.1/FVC was increased?p<0.05?and Cchord?p<0.01?was decreased significantly.The results showed that EEAV could improve the lung function,inhibit the level of inflammatory factors and improve the antioxidant capacity.It was concluded that EEAV,especially at 0.32 g·kg-11 dosage,had certain anti-inflammatory and antioxidant effects on cigarette smoke-induced COPD.In conclusion,the chemical constituents and anti-COPD activities of Allium victorialis cultivated in Jilin Province were studied in this thesis.The study will enrich the chemical constituents of Allium victorialis and expand its medicinal scope,provide scientific data and theoretical basis for further development and utilization of Allium victorialis L.
