| Objective(1)To understand the genotypes distribution characteristics and drug resistance of HIV-1 pairs of spouses positive family virus in some parts of guangxi,and to provide basis reference data for understanding the transmission laws and epidemic trends of HIV-1 strains in Guangxi;In the meantime,explore the relationship between the subtypes and the HIV-1drug-resistant,as to provide a reference for the better antiviral treatment.(2)To understand the HIV-1 cell tropism between different subtypes of gene in parts of HIV-1 pairs of spouses positive family,explore the relationship between the HIV-1 cell tropism and the process of disease infection.Methods Collected HIV-1 infections or patients by WB assary in HIV-1 confirmatory laboratory Nanning,Liuzhou,Guilin,Hezhou,Qinzhou,Guigang.A total of 77 HIV-1 pol region sequences were collected for HIV-1 genotype and drug resistance analysis.A total of 166 hiv-1 env sequences were used in the hiv-1 genotype subtype.Nucleic acid extraction by blood RNA extraction kit provided by Tian Gen company.Amplification of sequence fragments of HIV-1 pol and env genes by using In-house and RT-PCR.The amplification products are sent to Shanghai raw industry company forsequencing.Sequencing results were sequenced and processed with Sequencher software.Sequence alignment,shearing and translation of V3 loop amino acids by Bio Edit 7.0 Software.Analysis of the apical four peptide type,using MEGA software to build a phylogenetic tree.The pol region sequence was submitted to the HIV resistance database(https://hivdb.stanford.edu)of Stanford University for drug resistance mutation analysis.Using Geno2pheno(coreceptor)2.5 online tools to predict the use of accessory receptors,the amino acid sequence of V3 loop was compared with that of HXB2 reference strain,predicting the use of auxiliary receptors in the corresponding strains.Results A total of 77 patients were included in this study,age rang 25 to73 years old.50 cases(64.94%)were from the HIV-1 double positive family,27cases(35.06%)from HIV-1 single positive family.3 types of gene subtypes were found,there were CRF01-AE(89.61%),CRF07-BC(7.79%)and CRF08-BC(2.60%).Resistance detection showed that 8 samples had drug-related mutations.2 cases were double resistance to NRTIs and NNRTIs drugs.The incidence of drug resistance mutation was 10.39%.(2)A total of 166 cases came from HIV-1 double positive family were analyzed: It was found six subtypes of virulent strains,and there were 146 cases of CRF01-AE,9 cases of CRF07-BC,6 cases of CRF08-BC,1 cases of subtype B,2 cases of subtype C.In addition,2 cases of non-epidemic recombinant(URF)subtype 0107 were found.Four types of V3-tip motifs were found in this investigation,the main types are GPGQ(75.90%),the other three types were GPGR(10.27%)、GPGK(2.1%)、GPGH(1.4%)respectively.61.64% of the cases were predicted to use CCR5 as a co-receptor,38.36% of the cases were predicted to use CXCR4 as a co-receptor.Conclusion(1)In some parts of guangxi,the genotype of HIV-1 pairs of spouses positive family virus genotype was mainly popular with CRF01-AE,which mainly existed in heterosexual transmission population.The second is the CRF07-BC strain,which is mainly used in intravenous drug users.(2)The incidence of antiviral resistance of HIV-1 pairs of spouses positive virus in some areas of guangxi is moderately prevalent..(3)The presence of HIV-1 pairs of spouses positive family virus in both NRTIs and NNRTIs drug resistance.We should increase the detection and detection of drug-resistant strains in HIV/AIDS cases.(4)In some parts of guangxi,the top four peptides of the env gene V3 ring of the family virus env gene were mainly GPGQ,and the top four peptides from CRF01-AE,CRF07-BCand CRF08-BC were all GPGQ.(5)In some parts of guangxi,HIV-1 pairs of spouses positive family virus is mainly manifested as the non-syncytial induction type of macrophage,which is higher than other HIV/AIDS patients,which should cause concern. |
