?Objective?In this study,we cultured endothelial cells in vitro in different environments and observed the changes of endothelial cell function by hemodialysis to figure out the relationship between different factors and endothelial cell growth and find its possible mechanisms.?Method?1.Selected 40 healthy medical examiners randomly at the First Affiliated Hospital of Guangxi Medical University from June 2018 to November2018,in the mean time,77 maintained hemodialysis patients who get the hemodialysis(HD)were selected.Uesd their serum to intervene endothelial cells,detecting the proliferation rate and migration distance of endothelial cells.And detecting serum creatinine(Cr)and ?2-microglobulin,enzyme-linked immunosorbent assay(Elisa)was used to detect the content of interleukin-6(IL-6)before and after dialysis treatment;2.Human umbilical vein endothelial cells(HUVEC)were cultured with different concentrations of creatinine,?2-MG,IL-6,and IS to detect their proliferation activity and migration distance.3.After treatment with creatinine,?2-MG,IL-6 and IS for 24 hours,normalization was continued.After 24 and 48 hours,texted the proliferation activity of each group,and it was speculated whether or not the uremic toxin memory was established.4.Serum was intervened in MHD patients betweensingle hemodialysis(HD)treatment to detect proliferative capacity and migration ability,than detected serum creatinine,?2-microglobulin and IL-6.5.Analyze the correlation between creatinine,?2-MG,IL-6 and endothelial cell proliferation function,and calculate the linear regression equation.?Result?1.The cell proliferation ability of CKD patients was higher than that of healthy group.The cell proliferation ability of CKD patients before HD was higher than that of post-treatment cells,and the difference was significant(P<0.05).2.Healthy group and The difference of creatinine,?2-MG and IS in MHD patients was statistically significant(P<0.05).The levels of creatinine and?2-MG in patients with MHD were significantly higher than those after dialysis treatment.The level of IL-6 level after dialysis treatment was higher than that of dialysis.Before treatment,the difference was statistically significant(P<0.05).3.With the increase of creatinine and IS concentration,the inhibitory effects of creatinine,?2-MG and IS on endothelial cell proliferation were also gradually increased(P<0.05).However,IL-6 promoted the proliferation of endothelial cells,and its promoting effect increased with the increase of concentration(P<0.05).4.Creatinine,?2-MG and IS inhibited the healing ability of endothelial cells,and its effect was related to the concentration.The increase of IL-6 promotes the healing ability of endothelial cells,and its effect increases with the increase of concentration;5.Compared with the control group,the inhibition rate of creatinine,?2-MG and IS decreases with the intervention condition.The time increases and gradually decreases,while IL-6 promotes the proliferation rate.Pearson correlation analysis showed that the OD value of cells was positively correlated with creatinine(r=0.640,P<0.01)and ?2-MG(r=0.633,P<0.01),and IL-6.Negative correlation(r=-0.540,P<0.01),and the linear regression equation was y=0.000039a+0.002b-0.001 c.? Conclusion ? 1.Creatinine,?2-MG and IS inhibit the proliferation and migration of endothelial cells,IL-6 promotes endothelial cells and migration;2.Hemodialysis changes the function of endothelium cells by changing the concentration of uremic toxins(eg.creatinine,?2-MG and IL-6).
