| Objective: To observe the protective effects of Ginseng and Astragalus combination which are a part of Yangxin decoction on human umbilical vein endothelial cell, this study investigated the possible mechanism in the cell level to provide the theoretical basis for the prevention and treatment of UA disease.Methods: The rats were divided into Shenqi group and blank group randomly. Drug them for one week then takeblood , separate the serum, prepare medium including 2%, 4%, 6% Shenqi serum and blank serum. The HUVEC were seeded in culture plates and divided into 5 groups. They are 2% Shenqi group, 4% Shenqi group, 6% Shenqi group, blank serum group and model group. Overnight in an incubator at 37℃, put the medium into the right group then culture the cells in an incubator at 37℃for 24h.The model using H2O2 injury cells is similar to atherosclerosis model.Use electron microscopy to observe the changes in ultrastructure , detect OD values using MTT assay and calculate the rate of cell proliferation. Results:1.Electron microscope results showed that compared with the blank serum group, the cell shrinkage and cell gap was obviously enlarged in model group.With the increased concentrations, the cells were more liable to adherent grow in flat polygonal shape, intercellular closer to normal.2.The MTT results showed that compared with the blank serum group , the OD value was significantly lower in model group( P<0.01); compared with model group, the OD value of 2% Shenqi group has no significant increased( P>0.05), 4% Shenqi group was higher ( P<0.05), 6% OD Shenqi group was significantly higher ( P<0.01); the Shenqi group between the each two groups, the OD value of 6% Shenqi group is higher than the other two groups ( P<0.01) .3. The calculation of cell proliferation showed that the proliferation of model group was 50.94%, 2% Shenqi group group was 51.97%, 4% Shenqi group group was 57.99%, 6% Shenqi group group was 78.39%. Compared with model group, the cell proliferation rate of 2% Shenqi group has no significant increase( P>0.05), 4% Shenqi group was higher ( P<0.05), 6% OD Shenqi group was significantly higher ( P<0.01)Conclusion: The Ginseng and Astragalus combination serum has a clear preventive effect on the model using H2O2 injury HUVEC, which can promote proliferation of endothelial cells.Through Ginseng and Astragalus combination effect, they may uphold the system to improve endothelial dysfunction.To improve the abnormal vascular endothelial cells and prevent endothelial cell injury, that may play an important role in treatment of UA. |
