Expression And Significance Of Albumin/D-dimer And Long Non-coding RNA 00511 In Melanoma

ObjectThis study aims to investigate clinicopathological characteristics of melanoma,and explore potential prognosis and treatment indexes.MethodsIn the first part1.The subjects are 167 MM patients from the First Affiliated Hospital of Zhengzhou University.Then the clinicopathological features and follow-up data of patients were collected.2.Correlation analysis was utilized to study the correlation between clinicopathological parameters and overall survival(OS)/disease free survival(DFS).3.Single-sample t-test or?~2-test was used to explore the relationship between clinicopathological indexes and metastasis of tumor.4.ROC curve was used to study the hematological indexes related to metastasis,and regression tree analysis was used to determine the best cut-off value of albumin/D-dimer.5.Multivariate Cox regression analysis estimates the impact of multiple clinicopathological indexes on the incidence of metastasis.6.Kaplan-Meier curve was used to analyze the effect of single factor on OS,and Log-rank test was used to analyze the difference between survival rates.In the second part,1.Bioinformatics analysis and real-time polymerase chain reaction(qRT-PCR)were performed to measure the expression of LINC00511 in MM tissues and cell lines.2.Cell counting kit-8(CCK-8)was used to measure the effect of LINC00511 on cell proliferation.3.Flow cytometry was operated to test the effect of LINC00511 on cell apoptosis and cell cycle.4.Transwell assay was employed to determine the effect of LINC00511 on cell migration and invasion.5.Western-blot was performed to explore the relation of LINC00511 and some key protein molecules,including bcl-2,a-caspase-3,Akt,p-Akt and mmp-9.ResultsIn the first part,the results revealed that,after 15 years of follow-up,the median of overall survival time was 50 months,and the median of disease free survival time was 35 months.Moreover,by statistic analysis,a high level of albumin/D-dimer prognosis score(ADPS),albumin and low level of LDH,TNM stage,Brelow thickness,ulceration,and metastasis facilitated prognosis.Especially,ADPS was a metastatic biomarker for the prediction of MM at 341.01,cut-off value.In the second part,LINC00511 was highly expressed in MM tissues and cells.Also,high level of LINC00511 expression was positively associated with poor prognosis.Furthermore,LINC00511 silence could suppress the cell proliferation,cell cycle arrest,migration and invasion of A375 cells.Down-regulation of LINC00511could also induce the apoptosis of A375 cells.Then,we found that the expression of LINC00511 was negatively correlated with the expression of bcl-2,p-AKT and mmp-9,while silence of LINC00511 upregulate the expression of a-caspase-3 of A375 cells.Conclusion1.ADPS was a metastatic biomarker for the prediction of MM.2.Upregulated LINC00511 expression exerted oncogenic effects by promoting proliferation,migration,and invasion,and inhibiting apoptosis in melanoma cells.
