Objective(1)To explore whether there is a significant difference in the intestinal flora between the primary sj?gren syndrome(pSS)pSS and healthy people.(2)Further explore the specific differences in the structure of the intestinal flora of pSS.(3)Provide a corresponding reference for further research on the clinical basic research related to pSS and intestinal flora in the future.Methods Fecal samples of 14 women with pSS who met the inclusion criteria were collected,7 of whom were patients with qi and yin deficiency,7 with non-qi and yin deficiency,and 7 feces of healthy women aged 45-55.The bacterial total DNA of the collected fecal samples was extracted one by one,followed by PCR amplification,and high-throughput sequencing of the Illumina hiseq 2500 was performed on the V3-V4 region of the 16 SrDNA gene to obtain three sets of intestinal microbial bioinformation data,followed by inter-group Perform OTU analysis,structural diversity analysis,and significant difference analysis.Results(1)OTU mean comparison:There was no significant difference in the mean of the OTU between the healthy control group(n=7),the Qi-Yin deficiency type pSS(n=7)and the non-qi-yin-yin type pSS(n=7)(P>0.05).The complexity of the three sets of samples was analyzed by using the OTU Rank curve,the observed species index,the shannon index and the coverage index.The sequencing results showed that the measured data volume was complete,and the sequencing could almost completely reflect all the microorganisms of diversity information in the study.(2)There was no significant difference in the diversity of the three groups of intestinal flora.(3)Comparison of bacterial structure:Compared with the pSS patients,the healthy control group and the pSS patients were mainly composed of four fungi,Actinobacteria,Bacteroidetes,Firmicutes,and Proteobacteria.There was no significant difference between the four main groups in the two groups(P>0.05),but the range of fluctuations in the content of the bacteria in each group was significant.The difference between the healthy person and the non-qi-yin deficiency type pSS is Paraprevotellaceae and Ruminococcaceae(P<0.05).The bacteria belonging to the healthy and non-qi-yin deficiencytype pSS are Alistipes,Butyricimonas and Paraprevotella(P<0.05);the difference between the healthy person and the qi-yin deficiency type pSS is Mogibacteriaceae(P<0.05),and the genus of the healthy person and the qi-yin deficiency type pSS is Butyricicoccus(P<0.05);the species with significant differences between healthy people and pSS were Butyricicoccus_pullicaecorum and Lactobacillus_delbrueckii(P<0.05).(4)Analysis of the difference in the structure of pSS intestinal flora between different syndrome types:There are specific bacteria in the pSS patients with qi and yin deficiency.They are Chlamydiae,Fibrobacteres,Planctomycetes and Firmicutes,respectively.The specific phylogeny of pSS patients was Elusimicrobia,but they were not significantly different between the groups(P>0.05);the qi-yin deficiency type and the non-qi-yin-yin type pSS patients were compared with the genus Butyricicoccus.There was a significant increase in non-qi and yin deficiency patients(P<0.05),while Paraprevotellaceae was significantly increased in patients with qi and yin deficiency(P<0.05).Conclusion(1)There was no significant difference in gut microbiota diversity between pSS patients and healthy people,but there were significant differences in the proportion of specific bacteria in the flora structure.(2)The structure of the intestinal flora of pSS patients is characterized by the inhibition of inflammation and the maintenance of immune balance by the down-regulation of bacteria(Ruminococcaceae,Butyricicoccus,Alistipes,Butyricimonas and Paraprevotella).(3)The abundance of inflammatory flora in patients with qi and yin deficiency type pSS is higher than that of non-qi and yin deficiency type pSS patients(Chlamydia and Butyricicoccus),to a certain extent,the pSS patients with Qi and Yin deficiency syndrome are the disease.The main syndrome of the active period.
