Objective:To investigate the protective effect of peritumoral isolation band on skin burns caused by radiofrequency ablation in nude mice with breast cancer,and compare the protection effect in different solutions and injection methods.Materials and Method:In the first phase of the experiment,the purpose is to investigate the protective effect of peritumoral isolation band on skin burns caused by radiofrequency ablation in nude mice with breast cancer,and compare the differences in protection among different solutions.Forty five SPF ranked female BALB/c nude mice(5-week-old)were fed adaptively for one week.Then,the human breast cancer MCF-7 cells were implanted subcutaneously underneath the junction area of right forelimb and back in each nude mouse,thus MCF-7 tumor-bearing nude mice models were established.the mice were randomly divided into group A,group B and group C when the subcutaneous tumors grew to 8-12 mm in diameter.There are fifteen mice in each group.The nude mice in group A were injected with normal saline around the tumor,forming a peritumoral isolation band before radiofrequency ablation.The nude mice in group B were injected with 5% glucose around the tumor,forming a peritumoral isolation band before radiofrequency ablation.The nude mice in group C were not injected with any solution and no peritumoral band was formed before radiofrequency ablation.Then,the three groups of mice were operated percutaneous radiofrequency ablation guided by Color Doppler Ultrasound.The body weight and the tumor volume of the mice in the three groups were recorded before operation.The radiofrequency ablation time was recorded during the operation in the three groups.The wound surface area,wound surface temperature and wound healing time of the nude mice were measured in the three groups after operation.The survival of the nude mice was recorded,and the survival rate of the nude mice in the three groups was calculated.In the second phase of the experiment,the purpose is to compare the differences in protection effect among different injection methods.Thirty mice were randomly divided into group 1 and group 2.The mice in group 1 was injected with saline 5 ml,forming the peritumoral isolation band before radiofrequency ablation.The mice in group 2 was injected with saline 1 ml firstly,followed by continuous injection of saline using syringe pump with 1 ml/min during RFA process.The above data was also recorded.In addition,the data of thickness of the isolation band before and after the RFA were recorded.Count.Result:1,There was no statistical significance in the body mass,tumor volume and radiofrequency ablation time among the group A,B,and C(P > 0.05).2,The survival rates of nude mice in groups A,B,and C after radiofrequency ablation were 73.3%(11/15),80.0%(12/15),53.3%(8/15)respectively.The survival rate of nude mice in group A and B was higher than that in group C(P < 0.05).There was no significant difference in survival rate between nude mice in group A and group B(P > 0.05).The wound area of nude mice in group A and group B was significantly smaller than that in group C.The wound temperature in group A and group B was significantly lower than that in group C.The wound healing time in group A and group B was significantly shorter than that in group C(P < 0.05).However,there were no significant differences in skin wound area,wound temperature and wound healing time between nude mice in group A and group B(P > 0.05).3,There were no significant differences in body mass,tumor volume and radiofrequency ablation time between nude mice in group 1 and group 2(P > 0.05).4,The survival rates of nude mice in group 1 and group 2 were 73.3%(11/15)and 100.0%(15/15)respectively after radiofrequency ablation.The survival rate in group 1 was significantly lower than that of the group 2(P < 0.05).The wound area of the nude mice in group 1 was significantly bigger than that of the group 2(P < 0.05).The wound temperature of the nude mice in group 1 was significantly higher than that of the group 2(P < 0.05).The wound healing time of the nude mice in group 1 was significantly longer than that of the group 2(P < 0.05).There was a significant difference in the thickness of the isolation band between group 1 and group 2 before and after RFA(P < 0.05).The thickness of the group 1 was significantly reduced after surgery(P < 0.05),while the thickness of the group 2 was stable during the operation(P > 0.05).Conclusion:1,When nude mice bearing human breast cancer was operated radiofrequency ablation,the establishment of a peritumoral isolation brand could protect the skin from burn,but there was no significant difference in the protective effect between saline and 5% glucose.2,The method of continuous pumping is more effective than one-time injection,and the effect of skin protection is better and the effect is more stable.
