Effects Of Rice Wine On Rabbit Femoral Head Tissue Cells,Ultrastructure And VEGF Expression

Objective:To study the effect of rice wine on the tissue cells,bone microstructure and VEGF expression of femoral head in rabbits,and to explore the pathogenesis of alcoholic avascular necrosis of femoral head.Methods: Forty-four rabbits were randomly divided into three groups: Rice wine group(n = 20),saline group(n = 20)and control group(n = 4).The rabbits in the rice wine group were fed with rice wine by inserting the stomach tube three times a week.The same amount of normal saline was given in the 10ml/kg/d,saline group at the same time,while the control group was not treated with any treatment.The rats in the saline group were given normal saline in the same time.Before gavage(control group)and rice wine group,saline group was given intragastric administration for 12 months,and bilateral positive X-ray examination of hip joint was performed to find out the change of femoral head.The control group(n = 4)was selected as the control group(n = 4)before gavage,and the rabbits were randomly selected from the rice wine group and the saline group at 5 time points of 1,3,6,9 and 12 monthsrespectively.After X-ray examination,4 rabbits in each group were killed by air embolic method,and the rabbits in each group were killed by air embolization after X-ray examination.After gross observation,the changes of trabecular structure and ultrastructure were observed,and the number of osteoblasts and osteoclasts,the diameter of adipocytes and the ratio of empty bone lacunae were calculated,and the number of osteoblasts,osteoclasts,diameter of adipocytes and the ratio of empty bone lacunae were calculated.Immunohistochemical technique was used to observe the expression of VEGF.Results:(1)Gross observation: in rice wine group for 12 months,the sheen of femoral head was not uniform,and no obvious abnormality was observed at other time points.At each time point in saline group,no significant abnormality was found in the control group.(2)X-ray film:saline group,rice wine group and control group for 12 months,there were no obvious osteoporosis,cystic degeneration,collapse and necrosis of femoral head.(3)Observation under light microscope: after 3 months of gavage,the trabeculae of femoral head of rabbits in the rice wine group were gradually sparse and broken,and the bone tissue cells were disordered,while in the saline group,there was no obvious Abnormality at each time point in the control group.(4)Immunohistochemistry: after 6months of oral administration,the expression of VEGF in rice wine group decreased gradually,mainly in vascular endothelial cells and blood sinuses.At each time point in saline group,no significant abnormality was found in the control group.(5)The number of osteoblasts andosteoclasts: for 1 month,there was no significant difference between rice wine group and saline group and control group(P > 0.05).At 3,6,9,12 months after oral administration,the number of osteoblasts in rice wine group was lower than that in saline group(P < 0.05),and the number of osteoclasts in control group was higher than that in saline group and control group(P < 0.05).(6)The diameter of adipocytes,the percentage of empty bone lacunae and the expression of VEGF: there was no significant difference between the rice wine group and the saline group and the control group(P > 0 05).At 6,9,12 months after administration,the diameter of adipocytes and the percentage of empty bone lacunae in the rice wine group were higher than those in the saline group,and the expression of),VEGF in the control group was lower than that in the saline group and the control group(P < 0.05).(7)Observation of electron microscope: after 3 months of intragastric administration,the ultrastructure of femoral head in rice wine group was gradually destroyed,the bone cells were denatured and necrotic,and lipid drops were increased.At each time point in saline group,no significant abnormality was found in the control group.Conclusion:There were no significant changes in the ultrastructure of thigh bone,bone trabecular structure,bone tissue cells and microvessel density in rabbits treated with rice wine for a short time.For a long time,rice wine could cause degeneration and necrosis of femoral head ultrastructure,destruction of bone trabecular structure,disorder of bone tissue cells and decrease of microvessel density.With the prolongation of gastric perfusion time,it showed a trend of gradual aggravation.
