Partâ… : The expression of vascular endothelial growth factor gene transferred bone marrow stromal stem cells.Objective To study the expression of VEGF gene transferred BMSCs.Methods BMSCs were obtained from SD rat, the cultured BMSCs were transferred with the composite of pcDNA3.1-VEGF165 and Lipofecfamine regant in the ratio of 1/3 in vitro. The transcription and transient and stable expression of VEGF gene were investigate by using RT-PCR assay and immuno-histochemistry S-P method respectively. VEGF protein in culture medium were measured by enzyme linked immunosorbent assay(ELISA) method. The cell proliferation was detected by MTT method. Compared with the pcDNA3.1- transferred group and un-transferred group.Results the cell proliferation of the transferred BMSCs were similar to those of the control group. The transcription and expression of VEGF gene were found distinctly in gene transferred BMSCs after 48h of transfection and 4 weeks of culture. The control group were not found the transcription and expression.VEGF expression of the transferred BMSCs increased in 1 week,after that, the expression of VEGF gradually decreased.Conclusions 1. transferred by VEGF gene, the BMSCs can transcript and express the VEGF protein, ability of expression increased in 1 week, after that, the expression...
