Objective plasma long non-coding RNA H19?LncRNA H19?is inhibited after birth,and hypoxia can activate LncRNA H19.In the cerebral ischemia-reperfusion model of rats,LncRNA H19 level is significantly up-regulated after cerebral ischemia and hypoxia injury,which may be related to autophagy activation and apoptosis.However,the expression level of lncRNA H19 in patients with acute cerebral infarction remains unclear,and its clinical significance still needs further study.This study aimed to dynamic observating cerebral infarction patients LncRNA H19 expression level in different process,combined with clinical function score and bioinformatics methods,to explore the expression level changes in cerebral infarction and its clinical significance.Methods 20 patients with cerebral infarction admitted to the department of neurology of the first affiliated hospital of guangxi medical university from January 2018 to January 2019 were selected?age over 18 years old,initial onset,course of disease within 3 days,limb hemiplegia,consciousness?,and 20 healthy volunteers with age and gender match-were selected as the control group.Detect peripheral blood lncRNA H19 3 times in the process of its movement function recovery days 3,14 and 28 days.Healthy group at the same time detect lncRNA H19 once.Before each test,the National Institutes of Health Stroke score?National Institutes of Health Stroke scale,NIHSS?,short form FM motor function evaluation method?Fugl-Meyer?and the Barthel Index(Barthel Index,?BI?score was completed.Cerebral infarction group and the control group data compare used the two independent sample t-test,the spearman correlation analysis to investigate the patients after cerebral infarction,lncRNA H19 relative expression changes and neural function in patients with grade percentage,cerebral infarction group of each point in time with repeated measurement data analysis of variance analysis,data comparison between various time points using LSD analysis of variance.Lncrna2targetvv2.0 and starBase v2.0 were used to predict the lncRNA H19 target genes.The predicted target genes were analyzed through the GO and KEGG pathway enrichment analysis using the DAVID 6.8 website tool,so as to predict their basic bioinformatics functions and possible signal pathways.Results This study included 20 patients with cerebral infarction who met the inclusion criteria,including 14 males and 6 females.Infarcts occurred in the right hemisphere was 7 of the 20 patients,the left hemisphere was 9,and 4 in the brainstem.Most patients have traditional stroke risk factors.In terms of clinical manifestations,all the patients were sane,with mild to moderate degree of hemiplegia,and some patients were accompanied by partial numbness,central facial and lingual palsy,unclear speech and other manifestations of focal neurological defects.T-test between groups showed no significant difference in renal function,coagulation function,blood lipid and other indicators between the cerebral infarction group and the control group.During the observation period,the NIHSS score of cerebral infarction patients?12.2 5.83 vs 8.9 5.12 vs6.25 3.40,P<0.05?gradually decreased,and the FM score?55.70 12.04 vs 70.009.78 vs 78.50 8.73,P<0.05?and BI score?45.00 17.55 vs 60.00 15.04 vs 73.5013.58,P<0.05?gradually increased.The lncRNA H19 expression level of the patients was significantly up-regulated compared with the control group(2-??CTmethod for relative expression?averages and standard error?,the third day was 4.32±0.75;the 14th day was 1.80±0.24;the 28th day was 1.42±0.09;After the expression level of lncRNA H19 increased,it was gradually down-regulated with the extension of time and did not decrease to the normal level during the observation period.The percentage change in the above lncRNA H19 expression level was positively correlated with the percentage decrease in NIHSS score?R=0.447?,the percentage increase in FM score?R=0.463?,and the percentage increase in BI score?R=0.618??P<0.05?.Into the group of patients with cerebral infarction,averages and standard error 17.24±4.33cm3,Spearman correlation analysis found that cerebral infarction incidence after 3 days lncRNA H19 relative expression and cerebral infarction volume was significantly positive correlation?r=0.930,P<0.001?,cerebral infarction volume and days14 and 28 days after onset lncRNA H19 expression level had no significant correlation.Lncrna2targetv2.0 and starBase v2.0 online tools were used to predict 86 potential lncRNA H19 target genes.The enriched signal pathways include 55 signaling pathways we are concerned about,including apoptosis pathway,regulatory oxidative response,hypoxia response,cell stress,and P53signaling pathway.Conclusion LncRNA H19 may be a plasma biomarker for evaluating the severity of cerebral infarction.Changes in the expression of lncRNA H19 may be involved in the pathological process of neuronal apoptosis in the brain,and the increased expression concentration may be detrimental to the recovery of neurological function in patients.
