Objective To study the relationship between the expression of sonic hedgehog signal pathway and that of vascular endothelial growth factor(VEGF)in hypoxia human retinal pigment epithelial(hRPE)cells.Methods hRPE-19 were divided into normal control group and hypoxia control g roup.And the cells were cultured for 4h,8h,12h,24h,then applicat the fluores cence quantitative PCR(rt-pcr)to detect the level of mRNA in the supernatan of Shh and VEGF;The concentrations of Shh and VEGF was measured by enzyme-li nked im-munosorbent assay in the content by using Pearson correlation analysis.Results The control group showed a small amount of Shh,VEGF mRNA and protein expression.The expression in the hypoxia experimental group was significantly higher than that in the control group,and the difference was statistically significant(F=178.364,P<0.01).Moreover,there was a statistically significant increase in the inhibition group of cyclopamine(F =68.745,P<0.01).In addition,the expression of VEGF protein in the hypoxia group was positively correlated with the expression of Shh protein(r= 0.942,P<0.05).And the expression of VEGF and Shh mRNA waspositively correlated(r= 0.970,P<0.01).However,The results showed that there was no significant correlation in hRPE cells of cyclopamine suppression group(r = 0.915,P>0.05).Conclusion Shh signal pathway is involved in the expression of VEGF in hRPE cells under hypoxia.
