Improvement Of Antioxidant Capacity Of Sheep Bone Collagen Peptide(SBCP) And Collagen Peptide Chelated Calcium(CPCC) In Ovariectomized Rats

There is a certain relationship between estrogen deficiency,osteoporosis and the body's antioxidant ability.The aim of this study was to explore the sheep bone collagen peptide(SBCP)and the collagen peptide chelated calcium(CPCC)on osteoporosis(OP),and explore the antioxidant mechanism of OP.In order to study the effect of ovarian removal on the antioxidant ability of rats,the experimental model of ovariectomized rats was performed with the female SD rats as the subjects,the OP models were constructed,and the heart,liver,kidney,left femur and serum were collected.The results showed that the content of procollagen ty pe I N-terminal propeptide(PINP)and beta crosslaps(B-CTx)in the serum of group ovariectomy(OVX)increased significantly(P<0.01),the microscopic structure of the distal end of the left femur was significantly degraded,the protein concentration,T-AOC ability,T-SOD and GSH-Px activity of heart,liver and kidney significantly decreased(P<0.01),and MDA increased significantly(P<0.05),pathological changes in liver and kidney tissue slices compared with the group sham operation(SHAM).The conclusions showed that the OP model of ovariectomized rats was successfully constructed,and the removal of ovary not only resulted in osteoporosis,but also significantly decreased the antioxidant ability of organism.With O2-·,·OH and DPPH-free radicals was used as the index to judge the antioxidant activity of calcium preparation in vitro,and the antioxidant activity of IO-fig/mL SBCP,CPCC,maillard reaction products(MRP)and amino acid chelated calcium(AACC)were comparatively studied in vitro.The results showed that the scavenging effect of three kinds of free radicals was MRP>CPCC>SBCP,and the difference was very significant(P<0.01).Compared with the above three calcium preparation,the AACC of O2-·free radicals was significantly lower than CPCC(P<0.01)and no significant difference between CPCC and SBC?(P>0.05).The conclusions showed that MRP had very high antioxidant activity in vitro,and the antioxidant activity of CPCC in vitro was slightly better than SBCP.Group the ovariectomized rats,the effects of SBCP and CPCC on antioxidant ability in ovariectomized rats were studied by pumping the above subjects into the ovariectomized rats by the high,medium and low dosage groups after 8 weeks,compared with group SHAM,OVX and group estrogen(E2).The results showed that,compared with the group OVX,the high,medium and low dose groups of group OVX-E2,OVX-SBCP,OVX-CPCC,OVX-MRP and OVX-AACC all had increased the protein concentration,T-AOC ability,T-SOD and GSH-Px activity of the rat tissue homogenate,the improvement of the pathological state of the viscera,the microstructure of the femur and the decrease of MDA.With the increase of dosage,the effect was more obvious,and there was a tendency towards the group SHAM.Compared with the group OVX-E2,the effects of group OVX-SBCP,OVX-CPCC,OVX-MRP and OVX-AACC on the antioxidant indexes,microstructure and pathological changes of the femur in ovariectomized rats were not significant.In the group OVX-E2,the rats were irritable and had a mutual bite,while the other subjects had no such phenomenon.The conclusions show that SBCP and CPCC also contribute to the antioxidant ability of the body and have no other side effects while preventing and controlling OP.This study not only provides a theoretical basis for SBCP and CPCC to prevent and control OP's antioxidant mechanism,but also provides a safe and effective calcium preparation to prevent and control the occurrence of OP and improve the antioxidant ability of the organism.
