In this paper, the effects of ossein peptide on antioxidant capacity, indicators involved incollagen metabolism of skin and bone, bone strength in high-fat-diet KM mice were studiedby establishing a high-fat-diet inducing oxidative stress mice model, and1,2%ossein peptide,0.1%lipoic acid which was already known as antioxidant were supplemented respectively indiets.Materials and Methods:75male KM mice were randomly assigned to5groups(n=15),which respectively control group, high-fat-diet group, lipoic acid group,1%and2%osseinpeptide group. Blood was collected by removing the eyeball and all mice were sacrificed atthe end of16ws. Skins, livers and duodenums were taken to obtain homogenates for themeasurment of antioxidant capacity. Right femurs and No. five lumbar vertebras were takento do the analysis of biomechanical properties through texture analyzer and measurement ofbone calcium content. Skins and cartilages of thoracic vertebras were also collected for thecollagen measurement. Skins and left proximal femurs were dissected out and stored in Trizolfor the RNA extraction and genes amplification involved in collagen metabolism experiment.Results:1. Both high and low doses of ossein peptide could improve the imbalance ofredox state, decrease the rise of body weight, level of reactive oxygen species and content oflipid peroxide caused by high-fat significantly, increase the activities of antioxidase,strengthen body’s total antioxidant capacity. The antioxidant effect of high dose of osseinpeptide is equal with0.1%lipoic acid.2. Ossein peptide could markly improve the increasement of bone size and weaken ofmaximum load of femur and lumbar vertebra caused by high-fat diet, and could recover thereduction of bone calcium content in some extent.3. High-fat diet could reduce the content of hydroxyproline in skin and cartilagesignificantly, and ossein peptide could bring it back to normal, even higher than normal, andbetter than LA. The result of semi-quantitative RT-PCR showed the main ways which osseinpeptide influences collagen metabolism: not only can promote the synthesis of collagenthrough up-regulating the gene expression of prolyl4hydroxylase, but also can inhibit thedegradation of collagen through down-regulating the gene expressions of matrixmetallopeptidases and improve the ratio of relative mRNA levels of matrix metallopeptidasesand their inhibitors.Conclusion: Ossein peptide could markly improve the oxidatie stress state induced byhigh-fat diet, enhance the body’s antioxidant capacity, regulate gene expressiongs involved incollagen synthesis and degradation, maintain collagen level in skin and bone, and strenthenbone strength, increase the deposition of minerals.
