Preventive Effect Of Different Processed Products Of Crataegi Fructus On Mice With High Fat Diets Induced Obesity

Crataegi Fructus is one of medicinal material of our country in common use.It is a dried and ripe fruit of Crataegus pinnatifida Bge.or Crataegus pinnatifida Bge.var.major N.E.Br.It has a variety of processed products including Crataegi Fructus,fried Crataegi Fructus,charred Crataegi Fructus and carbonized Crataegi Fructus,and the property,function and indication of different processed products are not identical.It was found that Crataegi Fructus was widely used in health food with weight loss effect.Well,there is no systematic report on whether other processed Crataegi Fructus have the effect of losing weight.So,in this paper,the activity of different processed products was evaluated by enzyme activity test in vitro combined with rat model with nutritional obesity for the first time,and the changes of flavonoids fingerprints of different processed products were compared and analyzed.The major contents are summarized as follows:(1)Different processed Crataegi Fructus were prepared according to the traditional process by using Crataegi Fructus slices.The contents of catechin,epi-catechin,vitexin rhamnoside,hypericin,quercetin and isoquercetin were determined by HPLC.It was found that the contents of related components in Crataegi Fructus were 0.238%,0.029%,0.021%,0.393%,0.206%and 0.064%.After processing,the content of fried Crataegi Fructus decreased by 13.31%,44.22%,11.25%,41.90%,42.73%and 14.33%respectively;charred Crataegi Fructusdecreased by 25.05%?54.07%?31.74%?77.52%?94.74%and 68.53%,respectively;carbonized Crataegi Fructus decreased by 25.86%,82.03%,52.05%,82.72%,95.66%and 72.34%,respectively.(2)The effect of different processed Crataegi Fructus on pancreatic lipase activity was studied by pancreatic lipase activity test in vitro.Different processed Crataegi Fructus can inhibit pancreatic lipase to a certain extent,however,charred Crataegi Fructus has the best effect on inhibiting pancreatic lipase activity.The IC50 of charred Crataegi Fructus was 3.105 mg/mL.(4)The rats fed with normal diet were used as normal group,and obesity rat model induced by feeding SD rats with high fat diet.Orlistat,Simvastatin positive group and four processed product groups of Crataegi Fructus,fried Crataegi Fructus,charred Crataegi Fructus and carbonized Crataegi Fructus were set up.During the period,the body weight and food intake of rats were measured twice a week,and the general state observation was carried out after 30 days,at the end of the experiment,Lee's index was measured.Body fat rate and organ index were measured after sampling;determination of TG,TC,LDL-C,HDL-C content and pancreatic lipase activity in serum after blood coagulation;HE staining was performed on the perirenal and epididymal fat;oil red O staining was performed on the liver and pancreas.During the experiment,the whole mental state of the rats in each group was good,and there was no significant difference in the intake of food among the rats in each group.Compared with the model group,different processed products could significantly slow down the weight gain of rats,especially the charred Crataegi Fructus and carbonized Crataegi Fructus decreased by 31.2%,33.3%,respectively;charred Crataegi Fructus could significantly reduce the Lee's index of rats by 7.3%;can significantly reduce the accumulation of fat in rats;fried Crataegi Fructus and charred Crataegi Fructus could significantly reduce the accumulation of fat in rats,which decreased by 42.7%and 45.5%,respectively;fried Crataegi Fructus and charred Crataegi Fructus significantly decreased the liver index by 12.9%,13.9%,and the spleen index by 22.7%and 18.2%,respectively.Compared with the model group,charred Crataegi Fructus and carbonized Crataegi Fructus significantly decreased the content of TC,TG in serum of rats by 31.25%,29.46%and 38.4%,40.4%,respectively,and the content of LDL-C in rat serum decreased by 36.5%and 38.5%respectively;all Crataegi Fructus groups could significantly inhibit the activity of pancreatic lipase,and the highest reduction rate was 95.4%;Compared with the model group,the number of adipocytes in different processed Crataegi Fructus groups was smaller than that in the model group,especially the effect of charred Crataegi Fructus was the best.The diameter of perirenal and epididymal adipocytes decreased by 34.4%and 21.0%,respectively.The content of fat in the liver of each group was significantly decreased,especially the charred Crataegi Fructus and the content of fat decreased by 96.8%.The results of this paper show that:Different processed Crataegi Fructus have effect on anti-obesity,and it was found that charred Crataegi Fructus showed the best anti-obesity activity.It is suggested that charred Crataegi Fructus should be selected for health food or medicine of effect of anti-obesity.According to the effects of processing on six:flavonoids in Crataegi Fructus,flavonoids may have nothing to do with anti-obesity activity of Crataegi Fructus,and its degradation products maybe have the activity.It is suggested that the active components and their changing rules should be revealed by further research.
