Asarum sieboldii Miq.var.Seoulense Nakai,belongs to the plant of Aristolochiaceae is Shaoyin medicine in Korean Chinese folk medicine.It is used to treat cold,cough,headache and other symptoms.Asarum sieboldii Miq.var.Seoulense Nakai as aethnodrug,has pharmacological activities such as anti-bacterial,anti-inflammatory,anti-tumor,anti-pyretic and analgesic effects.The Korean Chinese medicine-Asarum sieboldii Miq.var.Seoulense Nakai,collected locally in Wang qing was used as the object of this study.The main goal of this study is to find compounds with antibacterial activity for the development of aethnodrug in China.The 95%ethanol reflux extract was segmentated using petroleum ether and ethylacetate separately.The antibacterial activity of each constituent was measured following.The extracts were isolated by various column chromatographic techniqies with silica gel column chromatography,hydroxypropyl dextran gel chromatography and high performance liquid chromatography.Eventually,fourteencompounds were isolated from Asarum sieboldii Miq.var.Seoulense Nakai including Aristolactam I(1);9-Methoxyaristolactam(2);7-Methoxyaristololactam IV(3);Aristolochicacid Iva(4);Asaramid II(5);Methyleugenol(6);(-)-Asarinin(7);(-)-Sesamin(8);(-)-Epipinoresinol(9);3,4-Dimethoxycinnamyl alcohol(10);(E,E,E)-2,4,8-decatrienoic acid isoutylamide(11a);(E,E,Z)-2,4,8-decatrienoic acid isoutylamide(11b);Aristololactam W(13);Estragole(14).Their structures were identified by spectra data including 'H-NMR,13C-NMR,TOF-MS/HR-ESI-MS,HMQC and HMBC.Among them,3,4-Dimethoxycinnamyl alcohol was isolated from Asarum plant firstly.According to the results of the antibacterial experiment,all compouds had antibacterial activity except for compounds 6 and 7,in addition follow the result of MIC,compounds 1,2,8,10,14 had obvious good antibacterial activity.Antibacterial activity of compounds 1 and 10 againsted drug-resistant bacteria like methicillin-resistant Staphylococcus aureus(3506),fungi like Candida albicans(7535)and gram-negativebacteria like Escherichia coli(1924)well,the minimum inhibitory concentration could reach to 8 ?g/mL.Compound 14 againsted six gram-positive bacterias,four Drug-resistant bacterias,E.coli(1924)and C.albicans(7535),especially for quinolone-resistant Staphylococcus aureus(3505)has the best activity,the MIC value could arrive at 16 ?g/mL.Compound 2 showed relative good activity against MRS A(3506),Staphylococcus aureus(RN 4220)and E.coli(1924),the MIC value could reach to 32 ?g/mL.The MBC value was generally less than 4 x MIC,which showed that compounds 1,2,5,8,10 and 14 had antibacterial effect and not easy to make bacteria produce drug resistance.The antibacterial activity of compounds 1,2,5,10 were reported for the first time.To analyze the chemical constituents of volatile oil from Asarum in seven areas,and to provide experimental basis for identification and utilization of Asarum from different habitats,volatile oil was extracted from Asarum by distillation method and the chemical constituents of volatile oil were analyzed and identified by GC-MS spectrometry in this experiment.The relative contents of these constituents were calculated by using peak area normalization method.We founded that the content of chemical constituents in volatile oil from seven habitats were different.The content of methyleugenol in volatile oil from Asarum which is the main pharmacologically active ingredients is the most in Yan Bian,it was reached at 47.58%,and the toxic compositions were latively low.Therefore,Asarum from Yan Bian,using as traditional aethnodug in China,had better therapeutic effect of antitussive,expectorant,sedative,analgesic compared to volatile oil from other producing areas.
