Objective:Study on the chemical constituents from Acer tegmentosum and the contents of major chemical components,study the pharmacokinetics in Rats.Methods:The air-dried stem back of A.tegmentosum were extracted with EtOH,and then extracted successively with CH2Cl2,EtOAc,and n-BuOH.The EtOAc extract was isolated and purified by multiple chromatographic techniques?normal and reverse phase silica gel column chromatography and performance liquid chromatography et al?.The NMR spectroscopy?1H-NMR and 13C-NMR?and their physicochemical properties were used for structural identification of monomeric compounds.The content of salidroside are determined by high performance liquid chromatography,The metabolic changes of the drug in rats were studied by using this indexes.Results:In this experiment,19 compounds were obtained from the ethyl acetate extract layer of A.tegmentosum,and identified by spectrum analysis as tyrosol?1?,6-carboxyl-?-?-catechin methyl ester?2?,methyl gallate?3?,protocatechoic acid?4?,gallic acid?5?,?+?-catechin?6?,quercetin-3-O-?-L-arabinofuranoside?7?,grayanoside A?8?,quercetin-3-O-?-D-arabinopyranoside?9?,eutigoside A?10?,?+?-gallocatechin?11?,salidroside?12?,?-?-epicatechin gallate?13?,?-?-epigallocatechin-3-O-gallate?14?,6''-O-galloylsalidroside?15?,3,3',4,4'-tetrahydroxybiphenyl?16?,quercetin?17?,quercetin-3-O-?-L-arabinopyranosyl-?1?2?-?-D-glucopyranoside?18?,sucrose?19?.Compound 2 was a new compound,compound 3 and 4 were the first time isolated from this plant,compounds 8,10,14 and 18 were isolated from the genus for the first time.The content of salidroside showed good linear range between 25.00?400.00 ?g·mL-1?R2=0.9996?,the average recovery rate of salidroside was 94.78%,RSD was 1.17%.The content of salidroside in the stem of A.tegmentosum was 1.3075%.The pharmacokinetic parameters of salidroside are:AUC0-t?1190.4 ±256.8?,AUC0-??1664.5 ± 456.0?,T1/2?5.9±0.4?h,Cmax?153.0±23.3??g·mL-1,Tmax?0.5 ±0.0?h,Vz?26.7±7.7?L kg-1,Clz?3.2±0.8?L·h-1·kg-1.Conclusion:In this experiment,19 compounds were obtained from the stem back of A.tegmentosum,and the main chemical constituents of A.tegmentosum was quantitatively analyzed,not only increased the understanding of this plant,but also enhanced the unnderstanding of the chemical composition of the genus.At the same time,a preliminary study on the pharmacokinetics of A.tegmentosum in vivo,this experiment providing a reliable pharmacokinetic theory for the development and application of A.tegmentosum.
