Effects Of Bu Yang Huan Wu Decoction With Different Concentrations Of Huang Qi On The Expression Of TLR4 And LOX-1 And The Mechanism Of Anti-atherosclerosis

Objective:Serum pharmacological methods were used to study the effects of Buyang Huanwu Decoction on TLR4 and its downstream dependent and independent pathways and LOX-1 expression,and to explore the mechanism of Buyang Huanwu Decoction in preventing and treating atherosclerosis.Method:1.Twelve experimental rabbits were randomly divided into blank group,high,medium and low dosage group of Chinese herbal medicine.Three rabbits in each group were intragastrically administered with saline and Buyang Huanwu Decoction with high,medium and low concentration of Huang Qi once a day for 7 consecutive days.One hours after the last administration,blood was collected from the heart,supernatant was extracted after centrifugation,and sterile filtered with a 0.22 micron disposable filler.The supernatant was stored in a refrigerator at-80 C for reserve.2.The third generation of human umbilical vein endothelial cells were resuscitated under sterile conditions and cultured at 37 C and 5%CO 2.3.Human umbilical vein endothelial cells were randomly divided into six groups:normal group,model group,western medicine control group,Buyang Huanwu Decoction group with high,medium and low dose of Huang Qi.LPS(1 ug/ml)was used to stimulate human umbilical vein endothelial cells,TLR4 and LOX-1 were activated,and different concentrations of Huang Qi Buyang Huanwu decoction containing serum and Ator vastatin were used to intervene.Cells were collected 24 hours later.Western blot was used to detect the expression of TLR4 and LOX-1 protein.Result:The expression of TLR4 and LOX-1 protein in the model group increased significantly 24 hours after LPS stimulation(P<0.01 compared with the blank group).After drug intervention,the expression of TLR4 and LOX-1 protein in the cells of the treatment groups decreased significantly,especially in the control group of Western medicine(P<0.01),and the expression of TLR4 and LOX-1 protein was significantly inhibited in the three traditional Chinese medicine groups(compared with the model group)(P<0.01).Buyang Huanwu Decoction with high concentration of Huang Qi has the best effect in three dosage groups.Conclusion:Buyang Huanwu decoction with different concentration of Huang Qi can inhibit TLR4 and LOX-1,which may be one of the mechanisms of preventing and treating AS.The expression of TLR4 and LOX-1 in Buyang Huanwu Decoction with high concentration of Huang Qi is the most obvious,which may be related to the mechanism of "Qi Neng Xing Xue".
