Effects Of Different Concentrations Of Astragalus Buyang Huanwu Decoction On PI3K/AKT Signaling Pathway

Objective:To study the effects of BuyangHuanwu Decoction on PI3K/AKT signaling pathway were studied by serum pharmacological methods,and the effects of BuyangHuanwu Decoction on atherosclerosis were further discussed.Method:1.Twelve rabbits weighing about 2 kg were randomly divided into four groups:blank group,western medicine control group,BuyangHuanwu Decoction high concentration Astragalus group,BuyangHuanwu Decoction medium concentration Astragalus group,BuyangHuanwu Decoction low concentration Astragalus group,three rabbits in each group.The blank group was given normal saline daily,and the other three groups were given BuyangHuanwu Decoction with different concentrations of Astragalus once a day for 7 consecutive days.On the last day,the blood was collected from the living heart and the supernatant was taken after centrifugation.2.Human umbilical vein endothelial cells(HUVECs)were cultured in vitro with ECM medium at 37 C,5%C02 and sterile conditions.3.Human umbilical vein endothelial cells were randomly divided into blank group,model group,western medicine control group,BuyangHuanwu Decoction high concentration Astragalus group,BuyangHuanwu Decoction medium concentration Astragalus group,BuyangHuanwu Decoction low concentration Astragalus group;intervention was carried out with corresponding serum containing drugs and atorvastatin,respectively,two hours later,except for the blank control group,five groups were stimulated with LPS(1 ug/ml);after 24 hours of intervention,cells were collected and proteins were extracted;Western blot was used to detect the expression of PI3K and p-AKT protein in each group.Result:After 24 hours of LPS stimulation,the expression of PI3K and p-AKT protein in the model group was significantly higher than that in the blank control group(P<0.01);the expression of PI3K and p-AKT protein in cells of each treatment group decreased significantly after the intervention of different drug serum,and the expression of PI3K and p-AKT protein in Western medicine control group decreased most significantly(P<0.01);the effect of BuyangHuanwu Decoction high concentration Astragalus group was the best among the three concentration groups.Conclusion:The serum containing Buyang Huanwu Decoction can inhibit the expression of PI3K and p-AKT protein activated by LPS stimulation,further inhibit the downstream inflammatory factors,thus having the effect of treating and reducing inflammation in atherosclerosis.Among the three groups of Buyang Huanwu Decoction with different concentration of astragalus,the Buyang Huanwu Decoction with high concentration of Astragalus has the best effect,which may be related to the traditional Chinese medicine theory of "Qineng Xingxue".
