Objective:In this study,the animal model of early diabetic nephropathy was prepared,and the therapeutic effect of Xiaoshen Recipe on early diabetic nephropathy(DN)was investigated.Meanwhile,the changes of inflammatory factors IL-1beta,IL-18,TNF-alpha and NF-?B in serum were analyzed to clarify its mechanism of action,so as to provide theoretical basis for the treatment of diabetic nephropathy by detoxification,dredging collaterals and protecting kidney.Method :112 healthy male Wistar rats weighing 200-220 g were randomly selected as the blank group after 5 days of adaptation in the animal room.Except the blank group,the other animals were injected with streptozotocin 60 mg/kg after 12 hours of fasting water.Blood sugar was measured 72 hours after fasting.The rats whose blood sugar was higher than16.7 mmol/L were regarded as successful hyperglycemia animal models.The successful rats were randomly divided into model group 10 and losartan potassium group 10.After 10 weeks of continuous administration,all animals were collected 24 hours urine to determine urinary protein,microprotein,creatinine and other indicators.Then the animals were anesthetized,abdominal aorta blood was taken,serum was separated,blood sugar,glycosylated serum protein,blood lipid,renal function and other biochemical indicators,TNF-a,IL-1 were detected.Inflammatory factors such as beta,IL-18.Result:1.Weight: Compared with the blank group,the body weight of the model group was significantly decreased(P<0.001).Compared with the model group,the body weight of the high-dose group,the middle and low dose groups,and the losartan potassium group were significantly increased(P< 0.01,P < 0.05).2.Kidney weight and kidney index: The kidney weight and kidney index of the model group were significantly increased(P<0.001).Compared with the model group,the kidney weight of the rats in the high,middle and low doses of the Xiaoshen Recipe and the losartan potassium group.The kidney index was significantly reduced(P<0.01,P<0.05).3.Blood biochemical indicators: Compared with the blank group,the GLU,GSP,TG,Scrand BUN of the model group were significantly increased(P<0.05,p<0.01,P<0.001),and there was no significant change in TC(P>0.05).Compared with the model group,the losartan potassium group decreased GSP,TG,Scr,and BUN(P<0.05,P<0.001),and had no significant effect on GLU and TC(P>0.05);The TG,GLU,GSP,BUN,TC and Scr in the middle dose group were significantly lower(P<0.05,p<0.01,p<0.001);the GSP and Scr in the low dose group of XiaoshenRecipe were also decreased(P<0.05).There was no difference in GLU,TC,TG and BUN(P>0.05).4.Urine biochemical indicators:Compared with the blank group,the Ucr,UP and UmA of the model group were significantly increased(P<0.05,P<0.001).Compared with the model group,the Ucr and UmA of the losartan potassium group were decreased(P<0.05,P< 0.001),UP no difference(P>0.05);UP,UmA,Ucr in the high,middle and low dose groups of Xiaoshen Recipe were significantly decreased(P<0.05,P<0.001).5.Inflammatory factors in serum: Compared with the blank group,the serum levels of IL-18,TNF-?,NF-?B and IL-1? in the model group were significantly increased(P<0.05,P<0.001);compared with the model group,losartan potassium The levels ofNF-?B and IL-1?in serum decreased(P<0.05,P<0.001),IL-18 and TNF-? did not change(P>0.05);serum NF-?B and IL in high-dose group of Xiaoshen Recipe-18,TNF-?,IL-1? content decreased significantly(P<0.05,P<0.001);serum NF-?B,IL-1? content decreased in the XiaoshenRecipe group(P<0.05,P<0.001),There was no difference in IL-18 and TNF-?(P>0.05).The serum IL-1? content in the low dose group of Xiaoshen Recipe was significantly decreased(P<0.001),but no significant effect on NF-?B,IL-18 and TNF-?.Impact(P>0.05).Conclusion:Xiaoshen Recipe can increase the weight of DN rats,reduce the kidney index,reduce blood sugar,blood lipid,reduce urinary protein,protect renal function,reduce the content of serum inflammatory factors IL-1beta,IL-18,TNF-alpha,and NF_kappa B.Inhibiting the expression of inflammatory factors is one of the mechanisms of Xiaoshen Decoction in the treatment of early DN.
