ObjectiveAfter 4 weeks,8 weeks,and 12 weeks of feeding on a high-fat diet,the pathological changes of glomeruli,changes in blood lipids,and changes in the expression of CaNA and NFAT2 proteins in C57BL/6J young mice were observed.The role and value of CaNA and NFAT2 protein in rat glomerular injury were investigated.MethodsThirty-six C57BL/6J male mice,all 3 weeks old,were randomly divided into 6 groups,each of which was 6 in number.Among them,the normal diet group applied standard feed to experimental animals,while the high-fat diet group used high-fat diet.The feeding time of both parties was 4 weeks,8 weeks and 12 weeks.The changes of body weight of 6 groups of mice were measured.The levels of serum triglyceride and serum total cholesterol were measured.The glomerular structure changes of experimental animals were observed by HE staining technique and Masson staining technique.Immunoblotting and immunization were performed.The histochemical technique was used to observe and detect the expression of CaNA and NFAT2 factors.ResultsIn the high-fat diet group,the weight of the young mice increased significantly compared with the normal group,and the serum triglyceride and total cholesterol levels were significantly increased.The HE and Masson staining results showed that compared with the normal diet group,the glomeruli of the glomeruli of the young mice in the high-fat diet group showed renal stenosis,renal sac fluid exudation,glomerular steatosis,collagen fibrosis,renal fibrosis,etc.Pathological changes.Immunohistochemistry and immunoblotting showed that the expression of CaNA and NFAT2 protein in glomeruli of the high-fat diet group was significantly higher than that in the normal diet group.ConclusionIn the glomerular injury of C57BL/6J mice caused by high-fat diet,the high expression of CaNA and NFAT2 protein may damage the structure and function of podocyte cytoskeleton protein,thereby infecting podocytes and making glomerular filtration.Over the disorder,proteinuria occurs;on the other hand,it may lead to glomerular hypertrophy,thickening of the glomerular matrix,glomerular fibrosis and ultimately kidney damage.
