ObjectiveTo investigate the effect of Kushen granule solution on P38MAPK?ERK related signaling pathway in rabbit ear acne model and its therapeutic effect on acne.MethodsAll 36 rats were randomly divided into 6 groups,6 rats in each group:blank control group,model control group,positive control group,and low concentration group of Kushen granule solution,Kushen granule solution concentration group,high concentration group of Kushen granule solution.With the exception of blank control group,the other five groups were established with the kligman method to establish the rabbit ear acne model.After successful modeling,the model control group,the positive control group,the Kushen granule solution high,middle and low groups were given physiological saline,fusidic acid cream,Kushen granule solution high,medium and low concentration topically,once a day for 14 consecutive days.The therapeutic effect was determined by HE staining microscope;The expression of TNF-?,IL-6 and IL-8 protein was detected by IHC method(Immunohistochemistry);The expression of P-P38 MAPK,P-ERK protein was detected by ELIAS(Enzyme linked immunosorbent assay);WB method(Western Blot)detection of P-P38 MAPK,P-ERK,P38 MAPK,ERK protein expression.ResultsCompared with the blank control group,the average optical density of TNF-?,IL-6 and IL-8 protein in the model control group increased,the content of P-P38 MAPK and P-ERK protein increased significantly,and the content of P38 MAPK and ERK protein decreased significantly,the difference was statistically significant(P<0.05).Compared with the model control group,the average optical density of TNF-?,IL-6 and IL-8 protein in each drug group and the positive control group decreased significantly,the content of P-P38 MAPK and P-ERK protein decreased significantly,the content of P38 MAPK and ERK protein increased significantly,and the difference was statistically significant(P<0.05).Compared with the positive control group,the average optical density of TNF-?,IL-6 and IL-8 protein in the high concentration group was similar,the protein contents of P-P38 MAPK,P-ERK,P38 MAPK,ERK were similar,the difference was not statistically significant(P>0.05),while the average optical density of TNF-?,IL-6 and IL-8 protein in middle and low concentration groups increased gradually,the protein contents of P-P38 MAPK and P-ERK increased gradually,the content of P38 MAPK and ERK protein decreased gradually,and difference was statistically significant(P<0.05).ConclusionHigh,medium and low concentrations of Kushen granule solution can decrease the expression levels of TNF-?,IL-6,IL-8,P-P38 MAPK and P-ERK,increase the levels of P38 MAPK and ERK,and intervene in the P38 MAPK,ERK pathway.To interfere with the formation of acne,which is effective for acne,the therapeutic effect increases with increasing concentration.
