Objective: The aim of this study was to establish a rat model of chronic high altitude disease(CMS)and research the mechanism of Apiumgrauens L.var.dulcedc extract intervention on CMS rat model.Methodss:(1)CMS rat model was established:50 SD rats were randomly divided into five groups,which were plateau model group(MG),plain control group(CG),nifedipine group(NE),Apiumgrauens L.var.dulcedc ethanol extract group(AG)and Apiumgrauens L.var.dulcedc water extract group(WG),and 10 rats in each group.Ten rats were fed under plain conditions for 45 days,40 rats were given 15 days after successful modeling,and the model needed to be fed under CMS conditions for30 days and measured pulmonary arterial pressure(PAP),C-reactive protein(CRP),vascular endothelial growth factor(VEGF),erythropoietin(EPO),angiotensinogen(AGT),apolipoprotein A1(Apo-A1)and islet-like growth factor-1(IGF-1).(2)To observe the morphological changes of pulmonary artery,heart,brain and lung tissues of rats in each group.(3)Forty SD rats were divided into five groups with 8 rats in each group: CG group,MG group,NE group,AG group and WG group.The serum differential changes of CMS rats were analyzed by metabonomics.Results:(1)Compared with the plain control group,the PAP of the plateau model group,alcohol group of Apiumgrauens L.var.dulcedc and water group of Apiumgrauens L.var.dulcedc and nifedipine group increased,with statistical significance(P< 0.05);compared with the plateau model group,the PAP of the nifedipine group,alcohol group of Apiumgrauens L.var.dulcedc and water group of Apiumgrauens L.var.dulcedc decreased,with statistical significance(P < 0.05).Comparing with the control group,the study found thatCRP,EPO,AGT,IGF-1,VEGF in rats respectively rise in the experiment of model group,nifedipine positive control group,alcohol group of Apiumgrauens L.var.dulcedc and water group of Apiumgrauens L.var.dulcedc(P<0.05),and Apo-A1 decreases steadily(P<0.05).Compared with model group,the study found that CRP,EPO,AGT,IGF-1,VEGF in rats decrease in theexperiment of alcohol group of Apiumgrauens L.var.dulcedcand water group of Apiumgrauens L.var.dulcedc(P<0.05)and Apo-A1 rises(P<0.05).(2)Under light microscope,Apiumgrauens L.var.dulcedc extract can improve the morphology of pulmonary artery,heart,brain and lung tissues in CMS rats.(3)After the intervention of Apiumgrauens L.var.dulcedc extract,lipid metabolism,sugar metabolism,amino acid metabolism and the metabolism of pyruvate,inositol,lactate and carnitine in serum of CMS rats have effected,suggesting that Apiumgrauens L.var.dulcedc extract can improve the internal environment disorder of chronic altitude disease model rats.Conclusion: Apiumgrauens L.var.dulcedc extract can significantly reduce the pulmonary artery pressure in CMS rats,improve the related serological indicators,target organ damage and metabolomics in CMS rats,and significantly postpone and improve the damage caused by CMS.
