Study On The Levels Of EPO, VEGF In Serum And EPO, EPOR In Tissues Of Rats With Chronic Mountain Sickness

Objective In order to understand the impact of EPO, VEGF on the pathologic process and to accumulate the important information of prevention and treatment in CMS, the serum EPO, VEGF levels and the expression of EPO, EPOR in the heart, liver, spleen and kidney tissues were detected in rats of CMS model.Methods The 49 healthy male Wistar rats were randomly assigned into the hypoxia group and the normoxic group. There were 24 animals in the hypoxia group and 25 animals in the another group. The rats of normoxic group were fed under the nature conditions in Xining for 30 days. The rats of the hypoxia group were fed in the hypobaric oxygen chamber for 30 days. The arterial blood was collected for whole blood cell analysis. The serum was separated and the levels of EPO and VEGF in serum were determined by ELISA. The expressions of EPO and EPOR in heart, liver, spleen and kidney tissues were detected by immunohistochemical assay.Results The Levels of HGB, RBC, WBC and HCT were significantly higher in the hypoxia group than those in the normoxic group, respectively. The mean pulmonary artery pressure was(66.06±17.36)mm Hg in hypoxia group, was significantly increased. The levels of PLT were(586.00±140.11)×109/L and(988.00±111.48)×109/L in hypoxia group and normoxic group, respectively, which showed that the PLT level in hypoxia group was lower than that in normoxic group(P<0.05). The serum levels of EPO, VEGF in hypoxia group were(13.29±2.55)ng/ml and(147.55±20.07)ng/ml, they were no significant difference with those in normoxic group accordingly. Also, the statistical difference was not found in the expressions of EPO and EPOR in the liver, spleen and kidney tissues between these two groups. But the expression of EPOR was lower in the heart tissue of hypoxia group.Conclusions It is concluded from the results of peripheral blood routine and pulmonary artery pressure determination that CMS animal models were established successfully by the method used in the study. The levels of EPO and VEGF in rats' serum and EPO, EPOR in rats' tissues of heart, liver, spleen and kidney did not significantly change in CMS rats. It was considered from the document literature that EPO and VEGF may play an important role in the acute hypoxia, but the pathologic mechanism of CMS is complex, there are many other factors might involved in it.
