Objective: To systematically study the antitumor activity and mechanism of Alpinia officinarum Hance against stomach cancer.Methods: 1)Transplanted MFC tumor animal model was established and mice were divided into six groups as follows: control group,model group,low-dose(15 mg/kg),middle-dose(30 mg/kg),and high-dose(60 mg/kg)groups of polysaccharides from Alpinia officinarum Hance and 5-Fu group.The mice were treated with polysaccharides from Alpinia officinarum Hance or 5-Fu for 14 days,except those of the control and the model group.The growth of tumor was observed.The tumor inhibitory rate,the spleen and the thymus index were calculated,respectively.The contents of IL-2、IL-6、IFN-γ、VEGF and PCNA in serum were detected by ELISA.2)Transplanted MFC tumor animal model was established and mice were divided into six groups as follows: control group,model group,low-dose(20 mg/kg),middle-dose(40 mg/kg),and high-dose(80 mg/kg)groups of TFAO and 5-Fu group.The mice were treated with TFAO or 5-Fu for 14 days,except those of the control and the model group.The growth of tumor was observed.The tumor inhibitory rate,the spleen and the thymus index were calculated,respectively.The contents of PCNA、MMP-9、VEGF、IL-1β、IL-6 and IL-17 in serum were detected by ELISA.3)CCK-8 assay was used to detect the effect of galangin on BGC-823 cell growth.Flow cytometry was used to detect cell apoptosis and cell cycle distribution.The enzyme activity of Caspase-3、Caspase-8、Caspase-9 was detected by kit assay.Western blot method was used to detect STAT3、Bcl-2、Bax、Caspase-3、Caspase-8、Caspase-9、CyclinB1、CyclinD1 protein expression in BGC-823 cell.Results: 1)Compared with the model group,tumor weight of mice decreased significantly(P < 0.05)in 5-Fu group,middle-dose and high-dose groups of polysaccharides from Alpinia officinarum Hance,the spleen index increased significantly(P < 0.05)in high-dose group of polysaccharides from Alpinia officinarum Hance while the thymus index of mice decreased significantly(P < 0.05)in 5-Fu group.Compared with the model group,the levels of IL-2 and IFN-γ in serum of mice increased obviously(P < 0.05)in 5-Fu group、middle-dose and high-dose group of polysaccharides from Alpinia officinarum Hance,the levels of IFN-γ in serum of mice increased obviously(P < 0.01)in 5-Fu group and high-dose group of polysaccharides from Alpinia officinarum Hance whereas the levels of IL-6,VEGF and PCNA in serum of mice decreased significantly(P < 0.01)in all administration groups.2)Compared with the model group,tumor weight of mice decreased significantly(P < 0.01)in 5-Fu group、low-dose、middle-dose and high-dose group of TFAO,the thymus index of mice decreased significantly(P < 0.05)in 5-Fu group and the spleen index decreased significantly(P < 0.05)in low-dose、middle-dose groups of TFAO.Compared with the model group,the levels of PCNA、MMP-9、IL-1β、IL-6 in serum of mice decreased obviously(P < 0.01)in all administration groups,the levels of VEGF in serum of mice decreased obviously(P < 0.01)in low-dose、high-dose group of TFAO and 5-Fu group,the levels of IL-17 in serum of mice decreased significantly(P < 0.01)in low-dose、middle-dose groups of TFAO and 5-Fu group.3)Galangin markedly inhibit the BGC-823 cell growth,accelerate the apoptosis,block cell cycle,increase cell Caspase-3、Caspase-8、Caspase-9 enzyme activity,upregulate the expression of Caspase-3、Caspase-8、Caspase-9、Bax and downregulate the expression of STAT3、CyclinB1、CyclinD1 and Bcl-2 protein.Conclusion: 1)Polysaccharides from Alpinia officinarum Hance has obvious anti-tumor effects.Its mechanism may be related to improving the immune function,inhibiting tumor angiogenesis and tumor cell prolifiration.2)TFAO has anti-stomach tumor activity,and mechanisms may include the inhibition of tumor angiogenesis,tumor cell prolifiration and cancer-associated inflammation.3)Galangin also has significant anti-stomach tumor activity,and its mechanisms may include the inhibition of STAT3-dependent apoptosis and cell cycle pathways.
