| ObjectiveTo investigate the expression level and significances of autophagy-related genes microtubule-associated protein light chain 3(LC3),Beclinl and matrix metalloproteinase-2(MMP-2)in the serum and peritoneal fluid of patients with endometriosis(EMs).To validate the mRNA expression levels of these three genes in the tissue of patients with EMs.To explore the relationship and relevance between these genes and endometriosis and its significances.To analyze the relevance between matrix metalloproteinase-2 and autophagy-related genes LC3 and Beclinl in the occurrence and development of EMs,so as to provide important theoretical basis for the pathogenesis of EMs,and to open up new horizons and provide effective markers for clinical treatment of EMs.MethodsThe serum and peritoneal fluid of 84 patients were collected in The First Affiliated Hospital of Qiqihar Medical University(Qiqihar,China)from March,2016 to March,2017.Among them,the serum and peritoneal fluid of 42 patients with EMs were regarded as experimental group,while the serum and peritoneal fluid of 42 patients without EMs were clarified as the control group.The levels of LC3,Beclin and MMP-2 in the serum and peritoneal fluid of patients with EMs and control subjects were detected by enzyme-linked immunosorbent assay(ELISA)assay.And the data were recorded for additional comparative analysis.In addition,qRT-PCR analysis was used to detect the mRNA expressions of LC3,Beclin and MMP-2 in the intima of the two groups.The dysregulated differences in the expression of these factors in the two groups were analyzed and evaluated.And we explored the correlation between the expression of LC3,Beclin,MMP-2 and endometriosis.Results1.The results of enzyme-linked immunosorbent assay(ELISA)showed that the levels of LC3 and Beclinl in EMs group were significantly lower than those in control group.2.However,the levels of MMP-2 in serum and peritoneal fluid from the EMs group were significantly higher than those in control group(p<0.05).The results of qRT-PCR showed that the mRNA expressions of LC3 and Beclinl in the endometrium of EMs patients were significantly lower than those in the control group,while MMP-2 was highly expressed in EMs patients(p<0.05).3.Matrix metalloproteinase MMP-2 was overexpressed in serum,peritoneal fluid and endometrium of patients with endometriosis,while autophagy-related genes LC3 and Beclinl were underexpressed.Matrix metalloproteinase MMP-2 was negatively correlated with autophagy-related genes Beclinl and LC3 in endometriosis.Conclusion:1.EMs patients showed the increased expressions of MMP-2 in the serum,peritoneal fluid and intima,and decreased level of LC3 and Beclinl.2.Matrix metalloproteinase MMP-2 is negatively correlated with the expression of autophagy-related genes LC 3 and Beclinl in endometriosis.3.This study explored the expression levels of MMP-2,LC3 and Beclinl in the patients with EMs in clinical.And it will help us to further clarify the pathogenesis of EMs and provide guiding significance for the clinical diagnosis and treatment of EMs.It enriches the study of the pathogenesis of endometriosis,helps to reveal the pathogenesis of endometriosis,and is expected to become a new target for the prevention,diagnosis and treatment of endometriosis. |
