Effect Of Zishentianjing Treatments On Endometrial HOXA-10mRNA Expression Of The Kidney Yin Deficiency Type Of IVF-ET Patients

Objective: To observe the effect of zishentianjing treatments on expression of HOXA-10 gene in kidney yin deficiency type IVF-ET patients and discuss the mechanism of the effect of zishentianjing treatments on endometrial receptivity.Methods: 120 infertile patients who were enrolled in the IVF-ET cycle and met the diagnostic criteria of Chinese and Western Medicine were enrolled in this study.Eighty patients with kidney-yin deficiency were randomly divided into observation group(40 cases)and blank group(40 cases),another 40 cases of simple fallopian tube factor for IVF-ET treatment of patients as the control group.The observation group in the first cycle of down regulation of menstruation on the 3rd to 14 days to take Erzhi tiangui granules,down regulation of menstrual 3 days to continue taking medicine to ovulation day,the control group and the blank group did not accept traditional Chinese medicine.Through patients with the informed consent to suck up a small amount of endometrial on the day of down regulation(usually 7 days after ovulation monitoring by ultrasonic monitoring),then load Eppendof tube in liquid nitrogen tank for storage,use RT-PCR to testing the expression of HOXA-10 mRNA in endometrium of patients on HCG injection day.Then to evaluate the changes of TCM syndrome score in two groups of kidney yin deficiency patients,and also evaluate the thickness,type,volume,uterine artery pulsatility index PI and resistance index RI of endometrium and serum hormone levels on HCG injection day.And compare the clinical pregnancy rate of these three groups.Results:(1)The score of syndrome of kidney yin deficiency in the observation group was significantly lower than that in the blank group(P <0.05),and the symptoms of kidney yin deficiency patients were improved;(2)The endometrial thickness and volume of HCG injection day of observation group were significantly higher than those in the blank group(P <0.05).There was no significant difference between the control group and the observation group(P> 0.05).The endometrial thickness and volume of HCG injection day of blank group were significantly lower than those of the control group(P <0.05).(3)The proportion of type A + type B of endometrial type in observation group was significantly higher than that in blank group(P <0.05),and the proportion was slightly higher in observation group than in control group,but the difference was not significant(P> 0.05);In blank group,the proportion of type A + type B of endometrial type was significantly lower than the control group(P <0.05);(4)The uterine artery pulsatility index PI and resistance index RI of endometrium in observation group on HCG injection day were lower than those in blank group,there was significant difference between the two groups(P <0.05);there was no significant difference between observation group and control group in uterine artery pulsatility index PI;but the RI in observation group was significantly higher than that in control group(P<0.05);Compared with the control group,the PI and RI of the uterine artery in the blank group were significantly increased on the HCG injection day(P <0.05)(5)The levels of serum estradiol(E2),serum luteinizing hormone(LH)and progesterone(P)in the three groups on HCG injection day were similar(P> 0.05);(6)Clinical pregnancy rate in the observation group was significantly higher than that in the blank group(P <0.05),but there were no difference between the control group and observation group(P> 0.05);the blank group's clinical pregnancy rate was significantly lower than control group(P <0.05)(7)The three groups all have the expression of HOXA-10 mRNA in the endometrium,but the expression in the blank group was the weakest,which was significantly different from the other two groups(P <0.05);the expression in the observation group was the strongest,which similar to the control group(P> 0.05).Conclusion: Zishentianjing treatments can effectively improve patient's endometrial receptivity,andthe clinical pregnancy rate increased significantly,and its mechanism of action may be related to the enhancement of endometrial HOXA-10 mRNA expression.
