Objective:The traits and microscopic characteristics of medicinal materials of the fruit of Lagenaria siceraria?Molina?Standl.var.microcarpa?Naud.?Hara were studied to further enrich the pharmacognosy data of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara,the contents of several major active ingredients were determined,the pharmacological activity of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara were studied,and the anti-diabetic substance basis was explored.The ethyl acetate extract of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara was isolated,purified and identified.Methods:The structure and powder characteristics of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara were observed and studied by microscopic observation methods.The water contents,ash contents,water-soluble extracts,alcohol-soluble extracts of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara were studied by using the method of"Chinese Pharmacopoeia".The contents of total triterpenoids,total polysaccharides and soluble polysaccharidewere were determined by ultraviolet spectrophotometry?UV?.The extraction of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara powder extracts were isolated and purified by useing the technology of ethanol reflux,silica gel column chromatography.The structures of these compounds were identified by modern analytical methods such as 1H-NMR and13C-NMR.The?-amylase inhibitory activity of the extracts was investigated and STZ-induced diabetes mellitus modelling method were used to develop the hypoglycemic pharmacological activity test of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara.Results:The traits and microscopic characteristics of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara and provides related picture data were described.The moisture,ash contents,water-soluble extracts,alcohol-soluble extracts,total triterpenes and total polysaccharides and other components of the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara were determined.The content of 7compounds was isolated and purified,and 3 structures were identified.It was found through the pharmacological experiments that the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara extract could reduce the activity of?-amylase and have a better therapeutic effect on the mouse model of type 2 diabetes inducted by STZ.Conclusion:A scientific basis for the identification of pharmacognosy for the fruit of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara was provided The foundation for further development and utilization of L.siceraria?Molina?Standl.var.microcarpa?Naud.?Hara medicinal materials was put forward.
