The Study Of IPO8 Promotes IL-6 Expression In Human Dental Pulp Cells In Vitro

Objective:.Importin 8(IPO8)that a nuclear transporters plays a important role in mediating the cytoplasm-to-nucleus transport of mature miRNAs.Preliminary in vivo studies have shown that the expression of IPO8 in specimens of pulpitis increased significantly,indicate IPO8 involved in regulation of pulpitis,but the mechanism is still unclear,By LPS stimulate the human dental pulp cells in vitro,the purpose of this article is by interleukin(IL)-6 expression to study IP-08 the role of human dental pulp cells.and its possible mechanism.Methods:HDPs were cultivated in vitro,according to different stimuli,set up LPS+HDPs group,LPS+HDPs+0.01ug/mlIPO8 group,LPS+HDPs+0.015ug/mlIPO8 group,the LPS+HDPs+0.02ug/mlIPO8 group and normal HDPs group,stimulation HDPs 12H and 24H,Real time-PCR and ELISA respectively to detect the expression of IL-6,to explore the exogenous IPO8 influence of the expression of IL-6 in HDPs,Western blot to detect the expression of IPO8 in LPS+HDPs group,to explore the expression oby Real-time PCR?ELISA respectively to detect the expression of IL-6,to explore the f IPO8 in HDPs.siRNA silencing IPO8,expression of IL-6 in HDPs.Results:Real time-PCR,ELISA and Western blot showed that than normal HDPs group,LPS+HDPs group the expression of IL-6 mRNA?protein and IPO8 protein significantly increased(*P<0.05),exogenous IPO8 stimulate HDPs further increase the expression of IL-6 mRNA and protein(*P<0.05),silent IPO8 after LPS stimulate HDPs,reduce IL-6 mRNA and protein expression(*P<0.05).Conclusion:IPO8 involved in regulation of HDPs and promote IL-6 expressed in HDPs.
