Objective:To study the expression of dentin sialophosphoprotein (DSPP)、bone sialoprotein (BSP) and alkaline phosphatase (ALP) of rat dental pulp cells, which were treated with lipopolysaccharide (LPS).Methods:Rat dental pulp cells were cultured by the method of tissue block in vitro and identified. The cells were treated with Porphyromonas gingivalis (Pg)LPS at concentrations of0.1ng/ml、1ng/ml、10ng/ml、100ng/ml and10000ng/ml. The effects of treatment were respectively examined by the expression of DSPP、BSP and ALP in rat dental pulp cells with the real time Rt-PCR after1、3、5days culture. The experimental data was statistically analyzed by spss17.0software package.Results:Under phase contrast microscopy, the adherent cells were found to be primarily typical fibroblasts, and part of the polygonal cells with protuberant cytoplasm. Real time Rt-PCR results showed that rat dental pulp cells expressioned higher mRNA of BSP、DSPP and ALP at concentrations of1ng/ml and10ng/ml, whereas the expression of BSP、DSPP and ALP mRNA were lower in groups of100ng/ml and10000ng/ml LPS. In1,3,5days, the expression of BSP、DSPP and ALP were gradually reduced in the cells which were treated with1ng/ml、10ng/ml、100ng/ml and10000ng/ml LPS; while the expression of DSPP^BSP and ALP was no significant difference in the group of0.1ng/ml LPS.Conclusion:Less concentration of Pg LPS promotes the expression of ALP%BSP%DSPP of dental pulp cells, but high concentration has inhibitory effect. With the extension of incubation time, the promoting effect weakens gradually, and inhibition effect is evaluated gradually.
