Expression Of IGFBP-1,Wnt4 And Bmp2 In Decidua Of Early Pregnancy And Their Relationship With Advanced Age And Embryonic Arrest

Objective:To investigate the expession of IGFBP-1,Wnt4 and Bmp2 in the decidua of normal early pregnancy and embryonic arrest and evaluate the relationship of Wnt4 and Bmp2 with decidualization during early pregnancy in human beings,and the their role in the pathogenesis of embryonic arrest.To explore the potential mechanism of advanced maternal age in embryonic arrest via age-stratified analysis of the expression of IGFBP-1,Wnt4 and Bmp2 in the decidua of embryonic arrest and normal early pregnancy.Methods:64 patients with embryonic arrest were enrolled into the study group.The study group was further divided into two age-stratified subgroups:No.1 sub-study group with 36 patients of age below 35,No.2 sub-study group with 28 patients of age equal to or over 35.The control group enrolled 70 women with normal early pregnancy who volunteered for abortion.The control group was also further divided into two age-stratified subgroups:No.1 sub-control group with 41 patients of age below 35,No.2 sub-control group with 29 patients of age equal to or over 35.Expressions of IGFBP-1,Wnt4 and Bmp2 in the decidua of study and control groups were detected by immunohistochemistry.Results:1.IGFBP-1,Wnt4 and Bmp2 expressed in the decidua of both normal early pregnancy and embryonic arrest,mainly in the cytoplasm and membrane of decidual cells.2.Expressions of Wnt4 and Bmp2 in the decidua of normal early pregnancy were both significantly positively correlated with the expression of IGFBP-1(p<0.01).Expressions of Wnt4 and Bmp2 in the decidua of embryonic arrest were both significantly positively correlated with the expression of IGFBP-1(p<0.01).3.Expressions of IGFBP-1,Wnt4 and Bmp2 were significantly stronger in the decidua of normal early pregnancy than their expressions in the decidua of embryonic arrest(p<0.01).4.Expression of IGFBP-1 in the decidua of No.l sub-control group was significantly stronger than its expression in No.2 sub-control group(p<0.01).Expression of Wnt4 in the decidua of No.1 sub-control group was significantly stronger than its expression in No.2 sub-control group(p<0.01).Expression of Bmp2 in the decidua of No.1 sub-control group was stronger than its expression in No.2 sub-control group and the difference was statistically significant(p<0.05).5.Expression of IGFBP-1 in the decidua of No.1 sub-study group was significantly stronger than its expression in No.2 sub-study group(p<0.01).Expression of Wnt4 in the decidua of No.1 sub-study group was stronger than its expression in No.2 sub-study group and the difference was statistically significant(p<0.05).Expression of IGFBP-1 in the decidua of No.1 sub-study group was significantly stronger than its expression in No.2 sub-study group(p<0.01).Conclusions:Expressions of IGFBP-1,Wnt4 and Bmp2 are lower in the decidua of study group,illustrating that Wnt4 and Bmp2 participate in decidualization of human beings,and the two factors' abnormal lower expression induce decidualization defect and embryonic arrest.Expressions of IGFBP-1,Wnt4 and Bmp2 in the decidua decrease as the age progresses.Expressions of IGFBP-1,Wnt4 and Bmp2 in the decidua of advanced maternal age are significantly lower,demonstrating that advanced maternal age has close correlation with decidualization defect.Advanced maternal age may induce abnormal lower expression of Wnt4 and Bmp2 which potentially result in embryonic arrest of advanced age patients.
