| Objective:To investigate the effects of IL-18 on retinal neovascularization and the expression of VEGF on mice.Methods:Fifty seven-day-old neonatal SD mice were randomly divided into five groups:control group,Nacl group,hyperxia experimental group, low concentration experimental group and high concentration experimental group.Nacl group,hyperxia experimental group, low concentration experimental group and high concentration experimental group exposed to (750±20)ml/L oxygen to establish models of retinopathy neovascularization for five days. The mice in low concentration experimental group and high concentration experimental group were respectively treated with intravitreal injection of interleukin-18 (1 u L) 10ng/μL and 50ng/μL. The mice in Nacl group intravitreal injection of Nacl (1μL). The animals were killed at the twenty-first day. Count the numbers of the endotheliocytenuclei of neovascularization which broke through the retinal inner limiting membrane by the HE-staining.The expression of VEGF and CD34 at layers of retina was detected by immunohistochemistry.Results:①The numbers of endotheliocyte nuclei of neovascularization so little in control group (1.40±0.70), but in hyperxia experimental group (14.7±2.26), low concentration experimental group (10.1±1.52) and high concentration experimental group (6.50±1.27), there are much more endotheliocyte nuclei and the low concentration experimental group more than high concentration experimental group.②The expression of VEGF was found at all layers of retina in each group, significantly increased in hyperxia experimental group (3.50±0.71) and Nacl group (3.70±0.48), relatively decreased in high (1.90±0.74) and low (2.60±0.52) concentration experimental group, and was to the lowest in control group.There were significant differences for low or high concentration experimental group compared with hyperxia experimental group and Nacl group(bothP<0.05), much more, the statistical difference was found between low and high concentration experimental groups (P<0.05).③The expression of neovascularization by CD34 was found at all layers of retina in each group, significantly increased in hyperxia experimental group (6.6±0.97) and Nacl group (6.9±0.11),relatively decreased in high (2.8±0.79) and low (3.8±0.79) concentration experimental group, and was to the lowest in control group.There was significant differences for low and high concentration experimental group compared (P<0.05).Conclusion:Interleukin-18 can inhibit the endotheliocyte nuclei proliferation and migration, effectively inhibit the retinal neovascularization. |
