Objective:Abnormality of tooth eruption such as the premature eruption,delayed eruption and im-pacted tooth is one of the common causes of malocclusion in clinic,which may affect the estab-lishment of normal occlusion,oral physiological function and appearance.It is still not clear about the relevant regulatory mechanisms affecting tooth eruption.It may bring positive research value and clinical significance by studying the relevant regulatory mechanisms of tooth eruption to prevent and treat abnormality of tooth eruption.At present,there has been no reports about the role of integrin ?5 on tooth eruption.This study aims to detect the histological localization and expression of integrin a5 during the tooth eruption,and to investigate the role of integrin a5 in the formation of hard tissue channels during the tooth eruption.Furthermore,it may provide a new theoretical basis for tooth eruption.It is expected to provide a innovative research direction for guiding the normal eruption of tooth in the future clinical work.Methods:Its content are divided into two parts.Part1:The purpose of this study was to quantify the expression of integrin ?5 during the tooth erup-tion.The first mandibular molar of the mouse was selected as the study subject to observe the initial period of tooth eruption(10 days after birth)and the middle period of tooth eruption(13 days after birth).The protein expression and mRNA level of integrin a5 were detected by immunohistochemis-try(IHC),immunofluorescence(IF),Western blotting and Real-time PCR respectively.Part2:The purpose of this study was to quantify the expression of OPG/RankL and osteoclasts in the formation of hard tissue channels during the tooth eruption.The immunofluorescence(IF)was used to observe the expression of OPG/RankL and osteoclasts in the formation of hard tissue chan-nels during the tooth eruption.Results:The result of part1:Integrin a5 showed positive expression patterns during the eruption of the first mandibular molar.The coronal bone tissue above the first mandibular molar began to be absorbed on the 13th day after birth.Polynuclear cells at the edge of the bone tissue indicated positive expression of integrin a5.The result of part 2:OPG/RankL and osteoclasts showed positive expression patterns during the eruption of the first mandibular molar.Integrin a5 and CD68+ showed co-expression.Conclusions:1.The results suggested that integrin a5 was positively expressed during the eruption of the first mandibular molars and might participate in the tooth eruption.2.The results suggested that OPG/RankL and osteoclasts were positively expressed during the erup tion of the first mandibular molars.And integrin ?5 and CD68+ showed co-expression.Therefore,we hypothesized that integrin ?5 may regulate osteoclasts through OPG/RankL signaling pathway so as to participate in the tooth eruption.
