IntroductionMetabolic syndrome(MS)is considered as a cluster of at least three of the five following conditions:central obesity,hypertension,glucose metabolism disorders,high serum triglycerides and low serum high-density lipoprotein cholesterol(HDL-c).MS is highly prevalent worldwide and in China.MS and its components interrelate to the risk of cardiovascular disease(CVD),stroke,and type 2 diabetes mellitus(T2DM),cancer,and increased all-cause mortality,which is a huge economic burden for individuals,families and public health care.The pathogenesis of MS,which has not been explained yet,is currently accepted to be influenced by various genes,environmental factors and their interaction.IGF2BP2 is reported to impact glucolipid metabolism,insulin secretion and insulin resistance.IGF2BP2 rs4402960,one of the most widely studied genetic locus,has uncertain biological function.Different studies on IGF2BP2 rs4402960 have inconsistent conclusions.Less association analysis has been reported on IGF2BP2 rs4402960 polymorphism with the longitudinal changes of MS components.Diversity exists in drug response among population with various genetic backgrounds.Pharmacogenomics helps medical decision.It is deserved to explore whether IGF2BP2 genetic variation affects the efficacy of medication on MS or its components.Objectives(1)To explore the distribution of IGF2BP2 rs4402960 polymorphism in a Chinese Han population;(2)To explore the association of IGF2BP2 rs4402960 polymorphism with the risk and progression of MS and its components;(3)To explore the effect of IGF2BP2 rs4402960 polymorphism on the efficacy of medication.MethodsStatistical analysis was based on Chinese Han population from January 2007 to December 2007 and a follow-up data of 5 years.Continuous variables were expressed with mean and standard deviations(SD)or Standard Error of Mean(SEM)and compared by Student'st test,paired t test,analysis of variance(ANOVA)with post hoc least-significant differences t test,or two factors analysis of variance.Categorical variables were presented with percentages and compared by ?2 test with Bonferroni correction when appropriate.Hardy-Weinberg equilibrium for rs4402960 was tested by?2 goodness-of-fit test,respectively in total population,controls and subjects with MS.Multivariate logistic regression and multiple stepwise linear regression was conducted to evaluate the risk factors and progression of MS and its components.ResultsIGF2BP2 rs4402960 polymorphism exists in Chinese Han population.In total subjects,IGF2BP2 rs4402960 polymorphism may relate to the risk of hypertension and MS.In males,IGF2BP2 rs4402960 polymorphism may relate to the risk of MS,increased waistline and hypertriglyceridemia.Allele T may associate with the progression of HDL-c reduction and hyperglycemia.Diuretics may have a better effect on SBP reduction in allele T carriers.ConclusionsIGF2BP2 rs4402960 polymorphism exists in Chinese Han population.IGF2BP2 rs4402960 polymorphism may relate to the risk of MS,especially increased waistline and hypertriglyceridemia in males.IGF2BP2 rs4402960 polymorphism may also associate with the risk of hypertension.Allele T may be associated with the progression of HDL-c reduction and hyperglycemia.Diuretics may have a better effect on SBP reduction in allele T carriers.
