LncRNA Analysis Of CD4+T Cells In Peripheral Blood Of Neurosyphilis Patients

Background:There is no gold standard for the diagnosis of neurosyphilis.Neurosyphilis was often misdiagnosed clinically due to its non-specific clinical manifestations,causing delay of the treatment and further damage.Dementia is a common clinical manifestation of neurosyphilis with high morbidity and mortality.Experiments on syphilitic animals suggested that CD4+T cells had involved in the immune response.Objective:To analyze the influence of Tpallidum on the clinical manifestations and laboratory results of neurosyphilis.Analyzing lncRNA expression of CD4+T cell in neurosyphilis patients.Methods:Neurosyphilis and dementia cases hospitalized in the department of neurology of Zhongshan hospital affiliated to Xiamen university from April 2009 to April 2019 were collected,and the clinical and laboratory characteristics were analyzed.The IncRNA expression profile of CD4+T cells of neurosyphilis in was analyzed,the effect of differentially expressed antisense lncRNA and related mRNA was analyzed.The expression of IncRNA ENST00000553061 was verified by qPCR.The full length of IncRNA ENST00000553061 was synthesized and connected with lentivirus vector then packaged.And the effect of overexpressed lncRNA-ENST00000553061 on cell proliferation was studied.Result:Compared with non-syphilis patients,neurosyphilis patients had more males,older,longer hospital stay,decreased peripheral blood lymphocytes,increased CSF white blood cells and elevated CSF protein.T.pallidum can t significantly stimulate proliferation of jurkat cells.Compared with non-neurosyphilis dementia,general paresis group has more males,younger,more hypertensive patients,lower clinical dementia score,higher total cholesterol and low-density lipoprotein and lower homocysteine.The lentivirus system of lncRNA ENST00000553061 was successfully established.And lncRNA ENST00000553061 is significantly down-regulated in CD4+T cells in neurosyphilis.Conclusion:The clinical and laboratory characteristics of neurosyphilis and general paresis were analyzed,suggesting age,gender,hypertension,diabetes,hospitalization time,cognitive level and CDR stage were related to neurosyphilis and general paresis.And decreased peripheral blood lymphocytes,increased CSF white blood cells,elevated CSF protein,higher total cholesterol and low-density lipoprotein and lower homocysteine are the clinical and laboratory characteristics of neurosyphilis and general paresis.In this study,we have established lentivirus system of lncRNA ENST00000553061,laying a foundation for further study on the effect of lncRNA on jurkat cells.We further studied the differentially expressed lncRNA of CD4+T cells in neurosyphilis and healthy controls,screening out the significantly down-regulated antisense lncRNA ENST00000553061 and verifying the clinical samples.These results provide some reference for the diagnosis of neurosyphilis.
