Objective:Atherosclerosis is a chronic,progressive and inflammatory disease,which is one of the main causes of death in patients with ischemic cardiovascular and cerebrovascular diseases.Abnormal activation of platelets and excessive pr-oliferation and migration of vascular smooth muscle cells?VSMC?play an i-mportant role in the formation and rupture of AS plaques.Methods:1.Effects of GETP and GEBH on platelet aggregation in hyperlipidemia rats in vitroThe platelet-rich serum was separated from the hyperlipidemia model of rats fed with high-fat diet.The platelet aggregation rate was measured within5 minutes after GETP and GEBH intervention in vitro by turbidimetry with A-DP as inducer.2.Effect of GETP,GEBH and GEBD on platelet aggregation in Hyperlip-idemia miceThe model of hyperlipidemia was established by intragastric administratio-n of fat emulsion in mice.After 14 days of GETP,GEBH and GEBD intraga-stric administration respectively,platelet-rich serum was separated.With ADP as inducer,the maximum platelet aggregation rate of GETP,GEBD and GEB-H in 5 minutes was deter-mined by platelet aggregator.3.Effects of GETP,GEBH and GEBD on PDGF-BB-induced proliferation model of A7r5 cellsThe proliferation model of A7r5 was induced by PDGF-BB.After adding different concentrations of GEBH,GEBD and GETP,the cell survival rate wa-s determined by MTS,The proliferation model of VSMC was induced by PD GF-BB.Flow cytometry analysis of the effect of GEBD on cell cycle in this model.4.Study on the Anti-atherosclerosis Effect of GEBH in RabbitsThe effects of GEBH on the level of TC,TG,HDL-C,LDL-C,TNF-?,PDGF-BB,VCAM-1,MCP-1 and P-selectin in serum were detected in rabbit AS mod-el induced by high-fat diet and intravenous injection of bovine serum albumi-nin ear margin.HE staining of thoracic aorta was performed.Results:1.Effects of GETP and GEBH on platelet aggregation in hyperlipidemia rats in vitroThe results showed that both GETP and GEBH could reduce the platelet aggregation induced by ADP in rats with hyperlipidemia in a concentration de-pendent manner.the IC50 of GETP is 0.53 mg/mL,95%confidence li-mit is0.49-0.59 mg/mL;The IC50 of GEBH is 0.15 mg/mL,95%confidence limit i-s 0.09-0.26 mg/mL2.Effect of GETP,GEBH and GEBD on platelet aggregation in Hyperlip-idemia miceThe results showed that GETP,GEBH and GEBD could reduce the platele-t aggregation induced by ADP in hyperlipidemia mice.3.Effects of GETP,GEBH and GEBD on PDGF-BB-induced proliferation model of A7r5 cellsThe results showed that after intervention by GETP?16?g/mL?and GEBD?10,20,40,80?mol/L?for 24 hours,the proliferation of A7r5 cells induced by PDGF-BB for 48 hours were inhibition,but GEBH could promote the prolife-ration of A7r5 cells.the percentage of G0/G1 phase cells increased significantl-y in GEBD group.4.Study on the Anti-atherosclerosis Effect of GEBH in RabbitsThe results showed that GEBH had obvious anti-atherosclerosis effect on rabbits,and could significantly reduce the contents of TC,TG,LDL-C,VCAM-1 in serum.Pathological sections showed that the thickness of aortic plaque in GEBH group was less than that in the mesentery,and the arrangement of smooth muscle cells in the mesentery was basically regular.The pathological changes in GEBH group were lighter than those in the model group.Conclusions:GETP,GEBH and GEBD can inhibit ADP-induced platelet aggregation in hyperlipidemia model in vivo or in vitro.GETP and GEBD can inhibit the proliferation of A7r5 cells induced by PDGF-BB.GEBD may inhibit the proliferation of VSMC induced by PDGF-BB by blocking cell cycle at G0/G1 phase.GEBH has obvious anti-atherosclerosis effect in rabbits.Its main function is related to regulating blood lipids and alleviating inflammatory cell adhesion reaction.
