Objective:The purposes of this study were to observe the effect of Gastrodiaelatablume phenolic compounds on the expression of VEGF-A and nerve repair after CIRI 7 days,14 days.Thus,clarify the effect of Gastrodia elata blume phenolic compounds on promoting angiogenesis and nerve repair.Methods: 1.By performing BSA gradient centrifugation method,brain microvascular endothelial cells(BMECs)are isolated and identified.Matrigel was used as cell scaffold and the tube number was used as angiogenesis index in vitro to investigate the effects of 3 #,4 #,8 # on capillary tube-like formation assay and select Gastrodia elata blume phenolic compounds which have angiogenic effects.2.Duplicating the rat MCAO/R model,intragastric administration was performed once a day for 7 days and 14 days with 3#,4# and 8#(the same below).Longa 5-points were used to neurological score at 2h,7days and 14 days after reperfusion.And neurological deficits scores was assessed after reperfusion 2h,and administered with samples 7days and 14 days,the ELISA assay was performed to investigate the secretion of VEGF in blood after rat administrated with 3#,4#,and 8#.The brain of rat were isolated when administration was performed for 7days and 14 days.Then immunofluorescence assay and Western Blot assay was performed to detect the expression of VEGF-A,NeuN and BDNF in brain cortex and hippocampus after rat administrated with 3#,4#,and 8#.Results: 1.The influence of 3#,4#,and 8# from Gastrodia elata Blume on capillary tube-like formation assay of rat primary BMECs: the number of tube in 3# group and 8# group were significantly increased,and 4# also shows an increased tendency.2.The influence of 3#,4#,and 8# on neurological function in rats: The neurological scores of rats after administration with 3#,4# and 8# in 7 days were decreased;the neurological scores were reduced significantly of rats group after reperfusion in 14 days.3.The effects of 3#,4# and 8# on the expression of VEGF-A in cerebral cortex and hippocampus of rats(immunofluorescence): after rats were treated with phenolic compound 3#,4#,and 8# in 7 days or 14 days,staining cells of VEGF-A in cortex of 3#,4#,and 8# group was increased significantly;after rats were treated with samples in 14 days,staining cells of VEGF-A in hippocampus of 3#,4#,and 8# group was increased significantly.4.Effects of 3#,4# and 8# on the expression of VEGF-A in the brain of rats(Western Blot): after rats were treated with phenolic compound 3#,4#,and 8# in 7 days the expression of VEGF-A in the brain of 4 # and 8# groups and after rats were treated with samples in 14 days,the expression of VEGF-A in the brain 8# group wincreased statistically(P<0.05),the expression of VEGF-A in the brain of 3# group at 7 days and 3#,4# goups at 14 days after treated with samples was increased.5.Effects of 3 #,4 # and 8 # on the secretion of VEGF in peripheral blood of rats: after rats were treated with phenolic compound 3#,4#,and 8# in 7 days,the secretion of VEGF in blood of 8# group was increased singnificantly,and 8# showed an increased tendency in VEGF after treated with samples in 14 days,4# showed also an increased tendency in the secretion of VEGF after treated with samples in 7 days and 14 days.6.The effects of 3#,4# and 8# on the expression of NeuN in rats cerebral cortex and hippocampus after reperfusion in 7days and 14days(immunofluorescence): after rats were treated with phenolic compound 3#,4#,and 8# in 7 days or 14 days,staining cells of NeuN in cortex of 3#,4#,and 8# group was increased significantly;staining cells of NeuN in hippocampus of 3#,4#,and 8# group was increased significantly after rats treated with samples in 14 days.7.The effects of 3#,4# and 8# on the expression of NeuN and BDNF in the brain of rats with reperfusion 7days and 14days(Western Blot): after rats were treated with phenolic compound 3#,4#,and 8# in 7 days or 14 days,the expression of NeuN of in brain of 3#,4#,and 8# group showed an increased tendency.After rats were treated with phenolic compound 3#,4#,and 8# in 7 days,the expression of BDNF of in brain of 4#,and 8# group and 3#,4#,and 8# group after rats treated with samples in 14 days was increased significantly.Conclusion:4# and 8# can promote capillary tube-like form.And Gastrodia elata blume phenolic compounds 3#,4# and 8# promote the angiogenesis and nerve recovery after CIRI.
