ObjectiveExplored mechanism of Gu ben fang xiao decoction prevent and treat of asthma by examining the effects of Gu ben fang xiao decoction on IL-33 and its signaling pathway-related factors ST2 Caspase3 Caspase7 and ERK1/2 in asthmatic mice.MethodsRandomly divided 24 female BALB/C mice into normal group model group and Chinese medicine group(Gu ben fang xiao decoction group).OVA combined with RSV attacked Mouse to construct a model of asthma persistence.With the HE staining to observed the pathological changes of lung tissue in each group.ELISA WB and qPCR were used to detect protein or mRNA expression levels of IL-33 and its pathway factor of sST2 caspase3 caspase 7 IL-5 ERK1/2 in the BALF serum lung issue bronchi.Results(1)Lung pathological results showed that compared with the normal group,the airway of the model group had obvious goblet cell hyperplasia,eosinophil accumulation,smooth muscle hypertrophy and others pathological changes.Compared with the model group,the pathological changes in the traditional Chinese medicine group showed significant alleviate.(2)BALF and serum results of ELISA showed that compared with the model group,the expression of IL-33 protein was significantly decreased in the BALF of the Chinese medicine group(P<0.05);the expression of sST2 protein in the BALF of the Chinese medicine group was increased(P<0.05);In the three groups’ serum the expression of IL-33 and sST2 were no statistical difference between the three groups(P>0.05).(3)Lung issue and bronchi results of WB showed that after intervention with traditional Chinese medicine the expression of IL-33 ERK1/2 protein in lung tissue was significantly lower than the model group(P<0.05);the expression of ST2 Caspase3 and Caspase7 in lung tissue was no statistical difference between the three groups(P>0.05);after intervene by traditional Chinese medicine the expression of IL-33 Caspase3 and ERK1/2 protein in the bronchi were significantly decreased(P<0.05);In the three groups’ bronchi the expression of Caspase7 was no significant difference(P>0.05).(4)Lung issue results of qPCR showed that after intervention with traditional Chinese medicine the expression of IL-33 ERK1/2 and IL-5 mRNA in the lung tissue of the mice were significantly lower than those in the model group(P<0.05);compared with the normal group,the expressions of ST2 mRNA in the model group was not obvious difference(P>0.05);compared with the model group,the ST2 mRNA in the Chinese medicine group was significantly higher than the model group(P<0.05);compared with the normal group,the expression of Caspase7 mRNA in the model group was significantly higher than the model group(P<0.05),It was no significant difference compared with the model group(P>0.05);the expression of Caspase3 mRNA was no significant difference(P>0.05).Conclusions(1)Gu ben fang xiao decoction can reduce lung inflammation in asthmatic mice,relieve goblet cell hyperplasia,smooth muscle hypertrophy,and inhibit eosinophil accumulation.It is indicated that Gu ben fang xiao decoction can alleviate airway inflammation and airway remodeling in asthmatic mice.(2)IL-33 is significantly elevated in this experimental model.Gu ben fang xiao decoction can significantly reduce IL-33 expression,ST2 protein expression,and competitive inhibition of IL-33 protein activity,suggesting that the role of Gu ben fang xiao decoction in preventing and treating asthma may be play by inhibiting the biological activity of IL-33.(3)Gu ben fang xiao decoction can alleviate the airway remodeling in asthmatic mice,which may be achieved by inhibiting the activation of IL-33/ST2/ERK1/2 signaling pathway and inhibiting cell over-repair.
